Purpose:The rat model of chronic heart failure was established by coronary artery ligation.Through the observation of the effects of Qiangxin Prescription on heart function,myocardial histomorphology,serum Ang II content,and the expression of myocardial MMP-1 and MMP-9 mRNA in rats with chronic heart failure.To explore the effect of ventricular remodeling in chronic heart failure rats and to clarify its mechanism.Methods:(1)The model of chronic heart failure in rats was prepared by coronary artery ligation and divided into model group,Shenfu group,Chinese medicine high,middle and low dose group and sham operation group according to weight.The hemodynamic changes were detected by the right carotid artery intubation.(2)The content of Ang-II in the serum of rats was determined by enzyme linked immunosorbent assay(ELISA).(3)The morphological changes of myocardial tissue and the degree of myocardial fibrosis in rats were observed by HE staining,Masson staining and transmission electron microscopy.(4)Detection of the expression of MMP-1 and MMP-9mRNA in myocardium by fluorescence quantitative PCR.To reveal the mechanism of Qiangxin vitality prescription on ventricular remodeling in chronic heart failure.Results:(1)Compared with the model group,hemodynamic indexes of Chinese medicine treatment group and Shenfu group were improved to varying degrees,and the improvement of middle dose group was the most significant,with statistical difference(P<0.01).(2)HE staining,Masson staining and transmission electron microscope were used to observe the myocardial tissue.It was found that the difference between the model group and the sham operation group was significant.The middle dose group had a prominent effect in improving cardiac muscle walking,cell morphology,and inhibiting myocardial fibrosis.(3)By detecting the content of Ang-II and the expression of MMP-1 and MMP-9 mRNA in the myocardium,it was found that the model group was significantly higher than the sham operation group,and the treatment group had different reduction,and the middle dose group had the most significant difference(P<0.05).Conclusion:Qiangxin vitality prescription can significantly improve the cardiac function of rats with chronic heart failure;it can inhibit the production of Ang-II in serum to prevent the process of myocardial fibrosis and cardiac hypertrophy,thus delaying the ventricular remodeling;it can reduce the expression of MMP-1 and MMP-9 gene to inhibit extracellular matrix reconstruction,further to the slowing of ventricular remodeling. |
