Gastric cancer(GC)is one of the most common malignant tumors in the world.It is characterized by high incidence,rapid progress and poor prognosis,which seriously affects human health.The high incidence of gastric cancer is particularly common in South Korea,Japan and China.According to China's urban cancer data report released by China's national cancer center in 2017,about 400,000 new cases of gastric cancer are diagnosed in China each year,and its trend is high.Although early gastric cancer can be removed by surgical resection,the five-year survival rate is up to 95%,but the postoperative survival rate is less than 30%.Currently,70% Chinese patients with gastric cancer are in the advanced stage when they are diagnosed.Therefore,the metastasis and invasion of gastric cancer are one of the important reasons for the low survival rate.With the continuous development of life science and medicine,the research on gastric cancer has achieved more results,but the underlying pathogenesis has not been clarified yet.Therefore,it has become an urgent problem to reveal the development of gastric cancer and the mechanism of its metastasis.The CT45A1 gene,as a member of the family of cancer and testosterone,has been found to be highly expressed in a growing number of malignant tumors,and the expression of CT45A1 is positively correlated with the prognosis of malignant tumors.Studies have shown that CT45A1 can play the role of transcriptional activator,which affects the expression of many tumor related genes from the transcriptional level,thus affecting the development of tumor from the molecular level.So far,the relationship between CT45A1 and gastric cancer has not been reported at home and abroad.Therefore,we ask scientific questions: 1.Whether CT45A1 promote the development and metastasis of gastric cancer?2.What is the molecular mechanism for promoting the metastasis of gastric cancer by CT45A1? In view of the above scientific problems,we propose :1.CT45A1 can promote the development and metastasis of gastric cancer.2.CT45A1 promotes the metastasis of gastric cancer by enhancing the expression of oncogene and inhibiting the expression of tumor suppressor genes.The research on this subject is carried out mainly from four levels: clinical data analysis,cell level,internal model and molecular level,so as to elucidate the relationship between CT45A1 and gastric cancer and its mechanism.This topic mainly studies CT45A1 genetic relationship with gastric cancer,and aims to reveal the CT45A1 in promoting the role of the occurrence of gastric cancer development and metastasis,and its molecular mechanism.So that they can provide new ideas and new strategies for the diagnosis and treatment of gastric cancer.First,through the data analysis of clinical samples,we found that with the increase of CT45A1 expression,the survival rates of gastric cancer patients showed a trend of decrease.It shows that CT45A1 expression level is closely related to the prognosis of gastric cancer patients is poor Secondly,in the cellular level,we have found that CT45A1 will markedly enhance their capabilities of clone formation of the gastric cancer cell lines,and it shows that CT45A1 promotes the gastric cancer cells' dedifferentiation and promotes the occurrence of gastric cancer.Through cell migration and invasion experiments,we found that the migration and invasion ability of gastric cancer cell lines with CT45A1 over-expression were significantly enhanced.Thirdly,we injected the control cells and the over-expression CY45A1 cells into the mice respectively.The results showed that the tumor in the experimental group was infiltrated,and lung metastasis occurred,indicating that CT45A1 could promote the metastasis of gastric cancer cells.Fourthly,through the detection of tumor related genes,it was found that CT45A1 activated the expression of multiple oncogenes such as ALDH1A3 and GFRA1,and reduced the expression of tumor suppressor gene PTEN.In addition,CT45A1 also activated the Erk signaling pathway of gastric cancer cells,thus promoting the development and metastasis of gastric cancer.In summary,CT45A1 gene promotes the development and metastasis of gastric cancer by activating oncogene and reducing the expression of tumor suppressor gene PTEN and activating Erk signaling pathway.The study on the role of CT45A1 in promoting the development and metastasis of gastric cancer and its molecular mechanism has not been reported at home and abroad.It is original and innovative,providing new ideas and new targets for the diagnosis and treatment of gastric cancer in the future.
