Excessive levels of reactive oxygen species in organisms can cause oxidative stress and oxidative damage to biological macromolecules such as proteins,DNA,and liposomes.And oxidative stress is closely related to physiological activities such as aging and a series of diseases such as cancer,cardiovascular diseases,and neurodegenerative diseases.Intracellular reactive oxygen over-oxidation of the protein thiol(RSH)side chain to generate protein sulfinic acid(RSO2H)is one of the important markers of this process.Thus protein sulfinic acid detection has important significance for the study of oxidative stress and related diseases,however its detection methods are still rarely.reported at present.Here,based on the nucleophilicity of sulfhydryl and sulfinic acid,we used the selective-relay strategy and found a-bromoacetophenones that can react with both sulfhydryl and sulfinic acids simultaneously.Then disulfides are synthesized and can selectively react with the sulfinic acid in the first step product.And we believe that research on detection of protein sulfination,can provide a powerful tool for the research and diagnosis of oxidative stress and related diseases.
