Objective: To study the effect of salvianolic acid A and C compatibility on inflammatory cytokines and oxidative stress reaction in HK-2 cells induced by HSA Methods :The cultured human renal tubular epithelial cells were randomly divided into 5 groups:control group,model group,salvianolic acid A group,salvianolic acid C group,salvianolic acid A and C group.After synchronization of 24 h,Except for the control group,the other group were added with HSA intervention for 24 h,48h,72 h,then the each treatment group was simultaneously added with the drug treatment.The enzyme linked immunosorbent assay method was used to detect the expression of MCP-1,CCL7 and IP-10.Immunofluorescence was used to detect MCP-1 and IP-10.M-CSF was detected by Western Blot.The levels of SOD,GSH and MDA were detected respectively.Result:1.The result of MCP-1,CCL7 and IP-10 1)Compared with the model group,the levels of MCP-1,CCL7 and IP-10 in the treatment group were relatively decreased in 24 h,48h and 72h(P<0.01).In the comparison of various treatment groups,the levels of MCP-1,CCL7 and IP-10 in the salvianolic acid A + C group were lower than those in the salvianolic acid A group and salvianolic acid C group(P<0.01).2)The positive expression of MCP-1 and IP-10 were green,and the expression were mainly localized in the cytoplasm of human renal tubular epithelial cells.2.The result of M-CSF Compared with the model group,the levels of M-CSF in the treatment group were relatively decreased in 24 h and 48h(P<0.05).In the comparison of various treatment groups,the levels of M-CSF in the salvianolic acid A + C group were lower than those in the salvianolic acid A group and salvianolic acid C group(P<0.05).3.Results of SOD,GSH and MDA Compared with the control group,the SOD and GSH levels were significantly decreased in 24 h,48h and 72h(P<0.05).Compared with the model group,the levels of SOD and GSH in the treatment group were relatively increased(P<0.05).the levels of SOD and GSH in the salvianolic acid A + C group were higher than those in the salvianolic acid A group and salvianolic acid C group(P<0.05).Compared with the model group,the level of MDA in the treatment group were relatively decreased in 24 h,48h and 72h(P<0.05).the level of MDA in the salvianolic acid A + C group were lower than those in the salvianolic acid A group and salvianolic acid C group in 24 h,48h and 72h(P<0.05).Conclusions: Salvianolic acid A and C drugs compatibility could reduce the expression of MCP-1,CCL7,IP-10 and M-CSF protein.Improve the vitality of SOD,increase the expression of GSH,and reduce the expression of MDA.The salvianolic acid A and C compatibility may improve renal fibrosis by decreasing the expression of inflammatory chemokines and antioxidation.
