| Objective:Chemically synthesize dopamine-hyaluronic acid material and construct dopamine-hyaluronic acid coating structure on dentin surface.This coating helps the dental pulp stem cells(DPSC)to efficiently and specifically adhere to dentin.This research will pave the way for forming the odontoblast cell layer in pulp regeneration.Methods: The dopamine-hyaluronic acid was synthesized by amidation reaction and characterized by some technologies,such as UV,FT-IR,and 1HNMR.The material was used to construct the dopamine-hyaluronic acid coating structure on dentin surface.The coating’s microcosmic morphology was investigated by scanning electron microscopy(SEM),and wettability was analyzed by contact angle detection.The change in coating thickness was examined by changing the concentration of this material.Then,DPSC and the dopamine-hyaluronic acid coated dentin model were co-cultured,and DPSC adhesion,growth and proliferation on that coated dentin surfaces were evaluated by SEM observation and MTT assay.Results:The dopamine-hyaluronic acid was successfully prepared,which has porous with the pore size of 110-130 μm.The water content is more than 90%.The dopamine-hyaluronic acid coating structure could be constructed on dentin surface.In a certain range,the coating thickness and the concentration of dopamine-hyaluronic acid material were positive correlation.The contact angle measurement confirmed that the dopamine-hyaluronic acid coating treatment of dentin could improve its hydrophilicity.Cell adhesion experiment showed that this dopamine-hyaluronic acid coating on dentin surface promotes DPSC to efficiently attach to the dentin,and exhibited good extension and growth state.The MTT results indicated that the proliferation of dentin + dopamine-hyaluronic acid coating group was superior to that of uncoated dentin group,and the difference was statistically significant(p<0.05).Conclusion: The dopamine-hyaluronic acid modified material was synthesized with stable chemical properties.The material could adhere closely to dentin,and form dopamine-hyaluronic acid coating structure on dentin surface.But also the dopamine-hyaluronic acid coating could induce DPSC-specific adhesion to coating-dentin.In addition,this coating had a promoting effect role in cell proliferation to some extent.In conclusion,the dopamine-hyaluronic acid can be used to construct coated dentin.It helps DPSC to efficiently adhere to dentin,and can pave the way for formation of stable odontoblast cell layer in pulp regeneration. |
