Objective: Objective to investigate the adhesion and proliferation of dental pul p stem cells on chitosan-fibrin scaffold.Methods:(1)The C57 rat dental pulp stem cells were isolated and cultured by modified enzyme digestion method.The cells were cultured for third generations,Identification of cells induced by adipogenic differentiation in vitro;(2)The preparation of chitosan-fibrin composite scaffold ma-terials,determination of the pore size and porosity;(3)The chitosan-fiber protein raw material composite scaffolds and the third generation of mouse dental pulp stem cells were cocultured and MTT assay was uesd to measure its 1~8 days of growth curve.(4)The morphology of the stent graft and the condition of cell adhesi-on,proliferation and extracellular matrix were observed under scanning electron mi-croscope.Results:(1)The C57 rat dental pulp stem cells were successfully isolat-ed and cultured,and the results were positive after osteogenic differentiation;(2)Chitosan-fibrin scaffold aperture is(105.32±22.10)um,the porosity is(87.714±1.276)%;(3)MTT assay shows that the proliferation growth curve of dental pulp stem cells is almost the same as the control group,which were cultivated on the chitos-an-fibrin composite scaffold material.Both of them are "S-shaped" curves.During the fourth day to eighth day,P<0.05,the proliferation rate of experimental group is significantly faster than control group,the difference is statistically significant;(4)On second day,cells were observed under scanning electron microscope of chitosan-fibrin composite scaffolds adhered tightly and grow in good condition.On fourth day,cells began to proliferate obviously,enlarge volume,be filled with rich extrace-llular matrix.On sixth day,cells covered the entire surface and void totally of ma-terials.Conclusion:(1)The dental pulp stem cells are easy to extract and show t-he biological characteristics of adipogenic osteogenesis;(2)As stated above,the c-hitosan-fibrin composite is a kind of scaffold material which is suitable for the ad-hesion and proliferation of dental pulp stem cells.
