Effect Of Solasonine On Cell Proliferation And Apoptosis In Human Gastric Cancer Cell Line MGC-803

objective:To observe the effects of solasonine on the proliferation of human gastric cancer cell line MGC-803 through in vitro experiment,and investigate that if they induce apoptosis by regulate the expression of Bax,Bcl-2,Caspase-3.Providing a New Vision for finding New drugs for gastric Cancer.Methods:Human gastric cancer MGC-803 cell lines were selected as research objects and treated with solasonine at concentrations of(5umol/L,10?g/m L,20umol/L,40?g/m L and 80?g/m L)for 24 h,48h and 72 h.Each concentration gradient was run in quintuplicate and MTT colorimetric assay was adopted to calculate proliferation inhibition rate of each group,and the ic50 concentration is obtained by calculation.A control group,20umol/L,40?g/m L and 80?g/m L were set according to ic50.And these concentration were used for the follow-up experiment processing 48 h.Western blot(Western blotting)was employed to detect the changes in the expressions of Bax,Bcl-2,Caspase-3.Result:MTT results showed that inhibition rates of solasonine at certain concentration(5umol/L,10umol/L,20umol/L,40umol/L,80umol/L)to the proliferations of human gastric cancer cell line MGC-803 for 24 h,48h,72 h were 3.81%,9.33%,11.14%,20.43%,81.15%;7.32%,11.81%,20.14%,30.21%,91.52%;14.22%,21.94%,30.24%,43.41%,96.71%,respectively.Inverted phase contrast microscope found that the cells in the negative control group grew well;And cells grew slowly,intercellular bridge disappeared,membranolysis,cell morphology irregularity were observed after the treatment of 48 h with various concentrations of solasonine.Western blot results showed that in a certain range,with the increase of solasonine concentration,the expression of apoptosis-related protein Bax,Caspase-3 increased,while the expression of apoptosis suppressor gene protein bcl-2 decreased,and the concentration of 80 umol/L was the most obvious.The concentration of 80 umol/L group was the most obvious.Conclusions:1.Solasonine at certain concentrations can inhibit in vitro proliferation of human gastric cancer cell line MGC-803,and its effect is in a time-and dose-dependent manner.2.Solasonine may induce apoptosis through infuencing the expression of Bax,Bcl-2 and Caspase-3.
