Objective: Through detectiving the expression levels of Breg/Th17 cells and related cell factors in peripheral blood of patients with NSCLC,analyzes the relationship of Breg/Th17 cells with clinical stage and chemotherapy in NSCLC patients,to explore the relationship of Breg/Th17 cells with occurrence and development of NSCLC and tneri potential role in tumorgenesis.Methods:There were 66 patients with NSCLC named as NSCLC group,and 20 cases of healthy subjects named as Healthy Controls.In the NSCLC group,12 cases in ?-? group,26 cases in ? guoup,28 cases in IV group,and there were 50 patients received chemotherapy drugs treatment.Fluorescent antibody CD19/CD24/CD27 mark Breg,CD4/IL-17 mark Th17.Though flow cytometry analysis and enzyme linked immumosorbent assay testing the proportions of Breg cells in CD19+B lymphocytes and the proportions of Th17 cells in CD4+T lymophocytes,and the expression level of related cell factors such as IL-10,IL-17 in peripheral blood of defferent groups,to explore their role in NSCLC.Ststistical analysis was performed by using the software 20.0.Results:1.The proportions of CD19+CD24+CD27+Breg cells in CD19+B lymphocytes in peripheral blood of NSCLC patients was significantly higher than those of Healthy Controls(16.66±7.98% VS4.41±2.56%),there was significant difference between two groups(P<0.05).The proportions of Th17 cells in CD4+T lymophocytes in peripheral blood of NSCLC patients was significantly higher than those of Healthy Controls(1.92±1.80% VS 0.86±0.62%),there was significant difference between two groups(P<0.05).2.There were significantly differences among diffenent TNM stages about of CD19+CD24+CD27+Breg cells in CD19+B lymphocytes in peripheral blood of NSCLC patients(P<0.05).The NSCLC patients in stage IV were significantly higher than those in stage ?-? and stage ?(8.36±3.58% VS 14.97±4.98% VS 21.80±7.83%).There were significantly differences among diffenent TNM stages about of CD4+IL-17+Th17 cells in CD4+T lymphocytes in peripheral blood of NSCLC patients(P<0.05).The NSCLC patients in stage IV were significantly higher than those in stage ?-? and stage ?(0.45±0.21% VS1.65±0.56% VS 2.80±2.38%).3.The proportions of CD19+CD24+CD27+Breg cells in CD19+B lymphocytes in peripheral blood of NSCLC patients who received chemotherapy drugs treatment was significantly lower than before(18.13±7.71% VS 12.56±5.52%),there was significant difference between two groups(P<0.05).The proportions of Th17 cells in CD4+T lymophocytes in peripheral blood of NSCLC patients who received chemotherapy drugs treatment was significantly lower than before(2.39±1.84% VS 1.46±1.03%),there was significant difference between two groups(P<0.05).4.The proportions of Breg cells in CD19+B lymphocytes was positively correlated with the proportions of Th17 cells in CD4+T lymphocytes in peripheral blood of NSCLC patients(r=-0.699,P<0.05).5.The expression level of IL-10 in peripheral blood of NSCLC patients was significantly higher than those of Healthy Controls(198.44±46.32% VS 116.42±35.34%),there was significant difference between two groups(P<0.05).The expression level of IL-17 in peripheral blood of NSCLC patients was significantly higher than those of Healthy Controls(34.22±10.21% VS 26.18±8.71%),there was significant difference between two groups(P<0.05).6.The proportions of Breg cells in CD19+B lymphocytes was positively correlated with IL-10 in peripheral blood of NSCLC patients(r=0.755,P<0.05).The proportions of Th17 cells in CD4+T lymphocytes was positively correlated with IL-10 in peripheral blood of NSCLC patients(r=0.355,P<0.05).Conclusion:1.Breg and Th17 cells can promote the occurrence and development of NSCLC.2.Breg cells was positively correlated with Th17 cells in peripheral blood of NSCLC patients,they have same effects on NSCLC.4.Breg and Th17 cells have same effect on the NSCLC may be concerned with their cell factors.
