Objctive To explore the expression and clinical significance of Th17 cells and IL-17 in the peripheral blood and tumor tissue in patients with Non-small cell lung cancer.Methods The NSCLC group was thirty cases of NSCLC patients. And the normal controls(NCs) was twenty cases of healthy volunteers. Flow cytometry(FCM) was performed to measure the level of Th17(CD4+IL-17+)cells in the peripheral blood, peritumor and intratumor tissue of NSCLC patients and the peripheral blood of healthy volunteers. Enzyme-linked immunosorbent assay(ELISA) was used to evaluate the concentration of IL-17 in the serum,peritumor and intratumor tissue of NSCLC patients and the serum of healthy volunteers. The relation of Th17 cells, IL-17 and clinical pathological features of NSCLC patients were analyzed.Result(1) The percentage of Th17 in the peripheral blood of NSCLC patients(4.05%±0.73%) were significantly higher than normal controls(2.12%±0.17%)(p<0.05). And the concentration of IL-17 in the serum of NSCLC patients(5.77±1.43) pg/ml were significantly higher than normal control(3.47±0.33) pg/ml(p<0.05).(2) The level of Th17 and IL-17 in theperipheral blood of NSCLC patients had positive correlation with the TNM stage(r=0.49 and 0.54, all p<0.05). The percentage of Th17 in the peripheral blood of NSCLC patients with stage III and IV(4.50%±0.45%) were higher than NSCLC patients with stage I and II(3.82% ±0.75%)(p<0.05). And the concentration of IL-17 in the serum of NSCLC patients with stage III and IV(6.63±1.20) pg/ml was higher than NSCLC patients with stage I and II(5.35±1.36) pg/ml(p<0.05).(3) In the peripheral blood of NSCLC patients, the percentage of Th17 had positive correlation with IL-17(r=0.47,p<0.05).(4) The percentage of Th17 in the intratumor tissue of NSCLC patients(5.15%±0.47%)were significantly higher than that of peritumor tissue(2.94%±0.31%)(p<0.05).And the concentration of IL-17 in the intratumor tissue of NSCLC patients(168.68±3.45) pg/ml was significantly higher than peritumor tissue(62.19±5.79)pg/ml(p<0.05).(5)The levels of Th17 and IL-17 in the intratumor tissue of NSCLC patients had positive correlation with the TNM stage(r=0.49 and 0.55, all p<0.05). The percentage of Th17 in the intratumor tissue of NSCLC patients with stage III(5.58%±0.21%) was significantly higher than NSCLC patients with stage I and II(5.03%±0.46%)(p<0.05). And the concentration of IL-17 in the tumor tissue of NSCLC patients with stage III(171.45±3.86)pg/ml was significantly higher than that of NSCLC patients with stage I and II(167.84±2.93) pg/ml(p<0.05).(6) In the intra-tumor tissue of NSCLC patients, the percentage of Th17 had positive correlation with IL-17.(r=0.64,p>0.05).Conclusions(1) The expression of Th17 and IL-17 in the peripheral blood and intra-tumor tissue of NSCLC patients are significantly higher than that of controls,and they have positive correlation with the TNM stage. This result suggests that Th17 cell and IL-17 maybe involved in the progression ofNSCLC.(2) In the peripheral blood and intratumor tissue of NSCLC patients,the Th17 have positive correlation with IL-17. This result suggests that the production of IL-17 is associatied with Th17 cell in the NSCLC. And this result also suggests that Th17 maybe take part in the progress of NSCLC by IL-17.
