In this study,the polysaccharide was extracted from Hericium erinaceus by water extraction and alcohol precipitation combined with degreasing,protein removal and small molecular impurities removal.The monosaccharide components were detected by infrared spectroscopy and gas chromatography.By establishing a tumor-burdened Lewis lung cancer mouse model of this Hericium erinaceus polysaccharide was studied in vivo antitumor activity and immune efficacy,and after chemotherapy combined with immune effect and synergies,as well as immune adjuvant and tumor vaccine joint tumor immune function.The results showed that hericium polysaccharide yield is 4.52%,the sugar content of purified polysaccharide was 88.5%,the purification of monosaccharide composition of polysaccharides mainly includes D-fucose(Fuc),glucose(Glu)and D-D-galactose(Gal),the mole ratio was 0.12:5.04:1.The infrared spectrum analysis of purified polysaccharides showed that it was a carbohydrate compound,which may contain acidic group and pyranose.Batches and subcutaneous inoculation of solid tumor with Lewis lung cancer way successfully established a tumor-burdened Lewis lung cancer tumors in mice model,adopt the way of irrigation to respectively for a tumor-burdened hedgehog fungus polysaccharides treatment in mice,made for kang(Irinotican,CPT-11)chemotherapy,hedgehog polysaccharides for kang synergy with the treatment,the treatment of 18 days in a row,treatment and tumor inhibition rate.The experimental results showed that the treatment of the polysaccharide of monkey can reduce the adverse reaction of the tumor to the body,and the synergistic chemotherapy of hericium polysaccharide could improve the survival rate of the tumor mice.The tumor inhibition rate of polysaccharide synergistic chemotherapy was significantly increased,which was positively correlated with the polysaccharide dose,and the tumor inhibition effect was higher than that of the chemotherapy drug CPT-11 alone.Moreover,polysaccharide has a certain role in regulating the secretion of cytokines and enhancing the immune function of the body,so as to assist the chemotherapy drug irinotecan to improve the anti-cancer effect.Using mitomycin C inactivated tumor cell preparation way of Lewis lung cancer tumor vaccine,using solid tumor Lewis lung cancer to extend and subcutaneous inoculation methods successfully establish a tumor-burdened Lewis lung cancer tumors in mice model,to a tumor-burdened Lewis lung carcinoma in mice tumor vaccine immune therapy.The experimental and immunotherapy experiments were conducted to investigate the treatment and tumor inhibition rate respectively,and the tumor inhibition effect was compared between the two experiments.Experimental results show that using mitomycin C inactivated Lewis lung cancer cells to do the regulation of tumor vaccine has certain level of cytokine secretion,enhance the body’s immune function and the effect of inhibiting tumor growth.The immune prevention experiment and immune therapy compared with the results of the experiment,tumor immune vaccine for prevention in advance can improve survival rate of mice a tumor-burdened relatively,the tumor inhibitory effect is obvious,and adding aluminum hydroxide in the tumor vaccine and polysaccharide has the adjuvant effect;After the tumor was inoculated,the therapeutic effect of tumor vaccine was not satisfactory,and the survival rate of the tumor mice was reduced,and the effect of adding aluminum hydroxide and polysaccharide without adjuvant was added in the tumor vaccine.
