| Hericium erinaceus is a valuable mushroom,which is not only delicious but also rich in nutrients and has wellness functions,and it has a long history of consumption around the world.Recent studies have shown that it contains a variety of active ingredients such as amino acids,polysaccharides,sterols,terpenoids,phenols and pyranoids,which have various biological activities such as anti-inflammatory,hypoglycaemic,anti-ageing,anti-tumour,hepatoprotective,and immunity-boosting.The food and medicine products made from Hericium erinaceus are widely welcomed by the market and have a broad market prospect for development in such areas.At present,a lot of researches has been conducted on the polysaccharide compounds and their bioactivities,but on the small molecule compounds.Therefore,it is necessary to study the small molecule compounds contained in Hericium erinaceus.Hericium erinaceus are widely cultivated in East Asia and Europe,and in China they are mainly produced in the northeast,but also in Zhejiang,Sichuan,Guangxi and Jiangxi provinces and regions.In this paper,the organic phase of ethyl acetate of the ethanolic extracts of the dried fruit bodies of Hericium erinaceus collected from Sichuan province was thoroughly investigated by recrystallization,silica gel column chromatography,sephadex column chromatography and PTLC,and the chemical components were isolated and purified.A total of 18 mono compounds were isolated and purified,and the structures of 13 compounds were elucidated by NMR(1D and 2D NMR),IR,HR-ESI-MS,etc,combined with the comparison with standards and TLC.They were:5α,8α-epidioxyergosta-6,22-dien-3β-yl linoleate(1),ethyl linoleate(2),linoleic acid(3),1,2,3-propanetriyl(9Z,9’Z,9"Z)tris(-9-octadecenoate)(4),herinece A(5)、herinece D(6)、herinece E(7)、(3,5,6,22E)-ergosta-7,22-diene-3,5,6-triol 3,6-dilinoleate(8),ergosta-4,6,8(14),22-tetraen-3-one(9),herinenone F(10),ergosterol(11),peroxyergosterol(12),3β,5α,6α,22E-ergosta-7,22-diene-3,5,6-triol-6-oleate(13).Among these elucidated compounds,compound 8 is a new compound,compounds 1,9,11,12 and 13 are known ergosterols or their derivatives,compounds 2,3 and 4 are long-chain fatty acids and their esters,and compounds 5,6,7 and 10 are phenolic compounds,which are typical compounds of Hericium erinaceus.Compounds 1,4,9 isolated from hericium reported for the first time.Based on the medicinal value of Hericium erinaceus,this paper investigated the anti-inflammatory activity of four phenolic compounds,herinece A(5),herinece D(6),herinece E(7)and herinenone F(10),obtained in this study,through an in vitro anti-inflammatory activity evaluation model,using the Elisa double antibody sandwich method to assess the compounds.The results indicated that the compounds herinece A(5)and herinenone F(10)had moderate anti-inflammatroy activity,with the IC50 of 78.50μM and 62.46 μM,respectively,while the compounds herinece D(6)and herinece E(7)did not have significant anti-inflammatory effects. |
