The distribution of Th9 cells and the expression ofOX40 in peripheral blood of breast tumor patientsObjective: To study the distribution of Th9 cells and the expression of OX40 in peripheral blood of breast tumor patients.Methods: To detect and analyze the expression of CD4+ T cells and OX40 and its relationship with clinical pathological parameters of invasive breast cancer(IBCa)by Flow cytometry.Results:(1)The percentages of Th9 cells in healthy and fibroadenoma patients peripheral blood were significantly higher than that in IBCa(P<0.01);(2)The expression of OX40 on CD4+ T cells,Th1 cells,Th2 cells,Th17 cells and Th9 cells of IBCa were significantly lower than those in healthy individuals or fibroadenoma patients(P<0.001,P<0.001,P<0.001,P<0.001,P<0.01).(3)The percentages of CD4+ T cells,Th2 and Th17 cells in the HER-2 negative group were significantly higher than that in the HER-2 positive group of IBCa(P=0.007,P=0.019,P=0.005);Conclusion: Th9 cells maybe participate in the immune response of breast tumors,and OX40 may play a role in maintaining the differentiation and function of Th9 cells.Part ? The expression of CD4,IL-9,OX40 and theirbiological significance in the invasive breast cancer tissuesObjective: To further study the expression of CD4,IL-9 and OX40 in IBCa tissues and their relationship with clinicopathological parameters.Methods: To detect the expression of CD4,IL-9 and OX40 in IBCa by immunohistochemistry.Results:(1)The percentage of CD4+ T cells was lower in lymph node metastasis than that without metastasis of IBCa(P=0.046);(2)The co-expression of CD4+ and IL-9 was found in IBCa(P=0.001,Kappa=0.294);(3)The co-expression of CD4 and IL-9 in ER or PR positive group of IBCa was higher than that in ER or PR negative group,respectively(P=0.022,P=0.014).Conclusions: The co-expression of CD4 and IL-9 was found in IBCa,which associated with ER or PR expression.Part ? Induction and differentiation of Th9 cells by OX40signal in breast tumorsObjective: To further study the induction and differentiation of Th9 cells induced by OX40 signal in breast tumors.Methods: To analyze the differentiation and function of Th9 cells under different culture condition in vitro.Results:(1)The levels of IL-9 secreted and OX40 expression by TEM or TCM cells in fibroadenoma than that in IBCa(P=0.009,P=0.003,P=0.025,respectively).(2)The expression of transcription factors PU.1 and the levels of IL-9 and their m RNA could induce increase by OX40 signal(P=0.002,P=0.007,P=0.036,respectively).Conclusion: OX40 signal can participate in regulating the differentiation of Th9 cells.
