Objective:To explore the whole genome DNA methylation modification of impulsiv-e aggression in patients with bipolar ? disorder.Methods:Using the Illumina human 450 k methylation chip technology to analyze the level of methylation of the whole blood genomic DNA samples of 5 cases of bipolar ? disorder patients with imp-ulsive behavior,5 cases of bipolar ? disorder patients without impulsive behavior.Using R software to preprocess the raw data.,then The select the difference methylation sites between sample groups.Finally,GCBI online analysis software was used to analyze the functional enrichment of different candidate genes.Results:(1)The samples of the attack group and the non-attack group were screened by methylation site,and the QC Report test obtained the various indicators to meet the quality requirements of Illumina.(2)Using beta as a measure of methylation,the beta density curve of all samples of the attack group and non-attack group tended to be consistent.(3)Using the Subset-quantile Within Array Normalisation(SWAN)algorithm to normalization the chip.The beta density curve tends to be consistent.,and the grouping meets the experimental design requirements.(4)Pooled t-test method was used to screen the difference methylation sites between groups,and there was a difference between the attack group and the non-attack group.(5)There was a difference in the distribution of the methylation sites between the attack group and the non-attack group on the chromosomes.(6)The differential methylation regions were classified by gene annotation classification and CpG island annotation classification,and the difference methylation area was distributed in 11 annotation regions.(7)The result of GO analysis shows that the differential methylation site participates in signal transmission and other functions.Pathway analysis is mainly for type I diabetes signaling Pathway.Conclusion: In this study,the total genomic DNA methylation level and the distribution of methylated DNA on the chromosome were different between the attackgroup and the non-attack group.Compared with the non-attack group,the distribution of the methylated regions was found in 11 regions classified by gene annotation and CpG island annotation.The difference methylation site between attack and non-attack group samples mainly engaging in biological functions such as signal transmission and dopaminergic neural synapses,chemokine signaling pathways and other signaling pathways.
