Purpose: By detecting the expression of circ-EIF4G3 in gastric cancer,to explore its role in the proliferation,migration and invasion of gastric cancer cells and its clinical value.Method: 1.Clinical research: The specimens of gastric cancer tissues and paracancerous gastric mucosa were collected from 64 patients with gastric cancer who were admitted to the Department of General Surgery,Nanjing Hospital Affiliated to Nanjing Medical University,from July 2015 to August 2016.We extracted RNA and reverse transcribed it to c DNA.q PCR was used to detect the expression of circ-EIF4G3 in cancer tissues and normal mucosa adjacent to cancer,and combined with the patient's data for statistical analysis.2.Cell function study: Two cell lines AGS and BGC-823 with the highest level of circ-EIF4G3 expression in gastric cancer cell lines were selected for si RNA knockdown.Circ-EIF4G3 was studied by CCK-8 and Trans-well and scratch experiments to explore the effects on cell proliferation,migration,and invasion.Result: 1.The expression level of circ-EIF4G3 in gastric cancer tissue was significantly higher than that in normal tissue adjacent to cancer(P <0.0001);2.The results of clinicopathological analysis suggest that circ-EIF4G3 has a certain reference value for the diagnosis of gastric cancer(AUC=0.7158),and the higher circ-EIF4G3 expression level indicates a shorter overall survival time(P=0.0191);3.In vitro experiments showed that circ-EIF4G3 can promote the proliferation,migration and invasion of gastric cancer cells.Conclusion: The high expression of circ-EIF4G3 promotes the proliferation,migration and invasion of gastric cancer cells.It has a certain reference value for the diagnosis of gastric cancer,and has the potential to be molecular marker for targeted therapy and prognosis evaluation of gastric cancer.
