Primary carcinoma of liver has high morbidity and mortality on the world.And it's one of the most common malignant tumors.The mortality rate of hepatoma in China is higher.In addition,the early symptoms of liver cancer are not obvious,and the later development is very rapid.These characteristics have raised the difficulty for the early treatment of liver cancer.Although the diagnosis and treatment of hepatocellular carcinoma in recent years have great development,but the survival rate of the patients with liver cancer is still not very ideal,in recent years on the deubiquitinase in more and more research in the development of tumor,deubiquitinating role involved in tumor formation,proliferation,metastasis and energy metabolism process.The activity of ubiquitination enzyme plays an important role in the development and development of liver cancer.PSMD14 is a member of the ubiquitination enzyme family,which regulates the de-ubiquitination of specific proteins to affect its stability and then affect the functional state of the cells.The abnormal changes of PSMD14 are not only directly related to the etiology of malignant tumors,but also have a close relationship with the development and prognosis of malignant tumors.It has been reported that PSMD14 can promote the proliferation,migration and invasion of tumor cells,but the specific mechanism has not been fully elucidated.In the experiment,immunohistochemistry and WB showed that PSMD14 expression level was significantly higher in HCC;PSMD14 promote cancer cell growth and metastasis;PSMD14 can interact with GRB2,and promote the stability of the protein through ubiquitin;PSMD14 affect GRB2's ubiquitination level.Conclusion:The high expression of PSMD14 in hepatoma can promote the growth and metastasis.PSMD14 can affect the protein stability of GRB2 by affecting the ubiquitination level of GRB2,thus regulating the development and development of liver cancer.
