Objective:To study the effect of taurine on Dimethyl benzene anthracene(DMBA-induced breast cancer in rats and its mechanism.Method:Establishment of Rat Breast Cancer Model: Thirty six-week-old female SD rats were placed in SPF level laboratory for one week and then randomly assigned to Blank group,Model group,and Ineintervention group,10 in each group.Rats in Model group and ineintervention group were treated with 15 mg/100 g of DMBA for one time,and 3% of taurine was added to the daily drinking water of ineintervention group rats;rats of Blank group were given 1 mL/100 g of peanut oil for one time.Gavage,all rats are free to drink and eat.Observation of the incidence of mammary tumors in rats every two days after administration.Enzyme-linked immunoabsorbent assay(ELISA): Detecting IL-6 and TNF-? levels in rat serumImmunohistochemistry: The expression of infiltrating lymphocytes(CD3,CD4,CD8,CD4/CD8)were detected in rat mammary tumor tissues.Results:Animal model construction results: During the entire animal observation blank group rats did not show any tumor around the mammary gland;compared with the model group,the mammary gland tumor latency in the intervention group was significantly increased(p<0.05),the incidence,the total tumor volume decreased significantly(p<0.05),there was no significant difference in the weight and volume of breast tumors(p > 0.05).Enzyme-Linked Immunosorbent Assay(Elisa): Compared with the blank group,serum IL-6 and TNF-? levels in the model group were significantly higher(p < 0.05).Intervention group rats serum IL-6 and TNF The level of-? was not significantly increased(p>0.05).Compared with the model group,the levels of serum IL-6 and TNF-? in the intervention group were significantly lower(p<0.05).Immunohistochemistry results: Compared with the blank group,the levels of infiltrating lymphocytes(CD3,CD4,and CD8)in the breast tumor tissue of the model group and the intervention group were significantly higher(p<0.05);compared with the blank group,The level of CD4/CD8 in the model group was significantly decreased(p<0.05),but there was no significant difference in the level of CD4/CD8 in the intervention group(p>0.05).Compared with the model group,the level of CD3 and CD4 in the breast tumor tissue of the intervention group And CD4/CD8 were significantly increased(p<0.05),but there was no significant difference in the level of CD8(p>0.05).Conclusion:Taurine has a significant inhibitory effect on DMBA-induced breast cancer in rats,and its mechanism may be through inhibiting the abnormal increase of serum TNF-? and IL-6 levels in rats,reducing the inflammatory response,and maintaining the normal body of rats.Immune mechanism,enhance immunity,promote TIL accumulation in rat breast cancer cells,enhance the body's immune amplification effect,direct killing of the original cancer cells,or kill tumors by secreting and mediating cytokines,thereby inhibiting tumors occur.
