Objective:To study the intervention and mechanism of taurine on breast cancer cells and nude mice with breast cancer.Methods:In vitro experiments:The group of group A breast cancer cells were cultured with L-15 cell culture medium supplemented with 10% fetal bovine serum and1% streptomycin double antibody.The taurine intervention group of group B Breast cancer cells were cultured with L-15 cell culture medium supplemented with 1% taurine,10% fetal bovine serum and 1% streptomycin double antibody.Drawing the breast cancer cell growth by curve and determining the breast cancer cycle by flow cytometry.In vivo experiment : Fifteen 5-week-old SPF nude mice were randomly divided into three groups.The 1 group of control group inoculated breast cancer cells and given normal drinking water.The 2 groups of intervention groups were inoculated with breast cancer cells and given 3% taurine drinking water.The 3groups of intervention groups were inoculated with 1% taurine medium for cultured breast cancer cells and given normal drinking water.The tumor volume was recorded every 2 days with a vernier caliper and Draw the tumor volume growth curve.40 days after treatment of nude mice,statistics of three groups of tumor mass and volume.The levels of estrogen receptor(ER),progesterone receptor(PR),human epidermal growth factor receptor 2(HER2)in human tumor were measured by immunoblotting.The expression of guanosine triphosphatase activating protein(p21ras)was detected by western blot.Results:In vitro experimental results:The cell growth curve results: the growth rate of breast cancer cells in group B was slower than that in group A.The flow cytometry test results: The proportion of G1 phase breast cancer cells in group B was(89.93±1.68%)(P<0.001).The proportion of G1 phase cells in group A was(84.9±2.08%)(P<0.001).The proportion of S phase breast cancer cells in group B was(66.54±2.17%)(P<0.001).The proportion of S phase cells in group A was(66.0±1.21%)(P<0.001).The proportion of G2 phase cells in group B was(6.68±0.15%)(P<0.001).The proportion of G2 cells in group A was(5.36±0.73%)(P<0.001).The proportion of M phase of breast cancer cells in group B was(19.49±0.93 %)(P<0.001).The proportion of M phase of breast cancer cells in group A was(17.59±0.35%)(P<0.001).In vivo experimental results:1?Nude mice breast cancer tumor model results,the tumor average quality of group 1 was(3.46±0.95g).The tumor average quality of group 2 was(2.02±0.75g)(P<0.05).Tumor inhibition rate was 41.6%.The average tumor mass of the group 3 was(1.35±1.23 g)(P<0.05).Tumor inhibition rate was 61.09%.The volume of the tumor in the group 1 was(3.06±0.004 cm3)greater than the group 2 of(1.79 ±0.006cm3)(P<0.05)and greater than the intervention group 3of(1.18±0.009 cm3)(P<0.05).2? The Western Blot result: Compared with the control group 1,the expression of ER protein was increased in group 2(P<0.05)and the expression of ER protein increased in group 3(P<0.05).The expression of PR protein in the group 2was increased(P<0.05)and the expression of PR protein in group 3increased by(p <0.05).The expression of HER-2 protein in group 2 decreased(P<0.05),and the expression of HER-2 protein in the group 3 decreased by55.4%(P<0.05).The expression of p21 ras protein in group 2 was decreased(P<0.01),and the expression of p21 ras protein in group 3 decreased(P<0.01).In summary,the expression of ER and PR protein in the two groups of taurine intervention group increased,and the protein expression of HER2 and p21 ras decreased.Conclusion1?Taurine has an inhibitory effect on the growth of breast cancer cells in vitro.It mainly by inhibiting the cell division cycle G1 and G2 phase,Mitotic completion of the expiration of DNA replication before the gap time and DNA replication is completed until mitosis begins.Suggesting that taurine may inhibit cell division by prolonging the G1 and G2 phases of breast cancer cells.Thereby inhibiting the growth of breast cancer cells.2?Taurine regulates the endocrine pathway by promoting the expression of ER and PR proteins and reduce the expression of HER2 and p21 ras protein,control the tumor cell division,proliferation link.Eventually achieving taurine inhibition in nude mice with breast cancer growth.
