Alveolar echinococcosis(AE)is caused by infection with the metacestode(larval)stage of the parasitic tapeworm Echinococcus multilocularis.The disease has a wide distribution in the world,a low cure rate,and brings serious economic burden on human society.One of the hallmarks of alveolar echinococcosis is tumor-like growth,invasion,and migration in the host after it has invaded the host,which is closely related to the self-renewal capacity and totipotency of germinal cells.Although the characteristics of the germinal cells in the E.multilocularis larvae have been demonstrated,the study of the cell cycle and its regulation mechanism has not yet been carried out.This dissertation mainly focuses on the cell cycle phase of germinal cells,and studies the growth and development mechanism of the E.multilocularis larvae.1.The characteristics of the cell cycle phase of germinal cells.The approximate time of the cell cycle of germinal cells was first detected by EdU single pulse.Then,the cell cycle phase of germinal cells is analyzed more precisely by EdU-BrdU dual labeling and EdU-pH3 dual labeling.Our data shows that the total cell cycle length of the germinal cells is 26 h,and the length of the four phase of cell cycle is 6.5 to 9.5 hours in G1 phase,8 to 10 hours in S phase,7 to 7.5 hours in G2 phase,and 1.6 to 2 hours in M phase?2.Effects of host hormones on the growth and development of E.multilocularis larvae.Larvae vesicles and protoscoleces are stimulated by host hormones in vitro cultivation,and then we observe the changes of cell proliferation in germinal cells.Our data show that the stimulation of the host hormones can promote the growth and development of the larvae vesicles and protoscoleces by inducing the Go phase cells to re-enter the proliferative state.In summary,the germinal cells have a short cell cycle and are able to undergo rapid division and proliferation,and tmaintain a stable amount in the metacestode larvae.Inducing G0 phase cells to re-enter cell proliferation is one of the important characteristics of the growth and development of E.multilocularis protoscoleces.This paper provides a feasible experimental method for the exploration of the characteristics of cell cycle,and it also provides a theoretical basis for the in-depth study of the cell cycle regulation mechanism of the germinal cells of the metacestode larvae.
