Objective:The expression of YAP(Yes associated protein),MST1(Mammalian sterile 20-like kinase 1)and CyclinD1 gene protein in NPC(nasopharyngeal carcinoma)tissues were detected,and its relationship with the clinicopathological factors,and the correlation between YAP and MST1 and CyclinD1 in NPC,and further explore the role of YAP in the occurrence and development of NPC.Methods:A total of 160 paraffin specimens of NPC tissue were collected from the pathology department of the Affiliated Hospital of Guilin Medical College from January 2009 to December 2012,the Median age of 47 years;40 cases of chronic inflammation of nasopharyngeal mucosa tissue samples were collected from the paraffin specimens of pathology department of the hospital from March 2016 to March 2017.The expression of YAP,MST1 and CyclinD1 in NPC and nasopharyngeal mucosa chronic inflammatory tissues were detected by Immunohistochemical staining method,and the prognosis of110 cases of NPC specimens was evaluated.The expression of YAP1 mRNA in patients with NPC and chronic inflammation of nasopharyngeal mucosa was detected by RT-PCR(reverse transcription Polymerase Chain Reaction).SPSS18.0 statistical software was used to analyze the results of above experimental,Person?~2 test was used to analyze the differential expression of YAP,MST1,CyclinD1 in NPC and chronic inflammation of nasopharyngeal mucosa tissues,and analyze the relationship between the three and the patients' sex,age,pathological classification,lymph node metastasis,clinical stage,distant metastasis and recurrence.The correlation between the three of YAP,MST1 and CyclinD1 were analyzed by Spearman's correlation coefficient for ranked data.One-way analysis of variance was used for analysis of the differential expression of YAP1 mRNA in NPC and chronic inflammation of nasopharyngeal mucosa tissues.Kaplan-Meier was used for evaluate the relationship between the expression of YAP,MST1,CyclinD1 and the overall survival time of tumor patients after radiotherapy and chemotherapy.The difference was considered statistically significant with p<0.05.Results:(1)The positive expression rate of YAP,CyclinD1 and MST1 protein in NPC tissues was 84.375%,92.5%and67.5%respectively.The positive expression rate of YAP,CyclinD1 and MST1 protein in the chronic inflammation of nasopharyngeal mucosa was 52.5%,57.5%and 92.5%,The difference was statistically significant(p value was0.000013,0.00000002,0.002 respectively).(2)The high expression of YAP and CyclinD1 was significantly correlated with pathological type(p value was0.028,0.005 respectively),It was not related to the patients' sex,age,lymph node metastasis,clinical stage,distant metastasis and recurrence(all p>0.05);the low expression of MST1 was not related to the patients' sex,age,pathological type,lymph node metastasis,clinical stage,distant metastasis and recurrence(all p>0.05).(3)the expression of YAP was positively correlated with CyclinD1(r=0.270,p=0.001),not related to the expression of YAP and MST1(r=0.069,p=0.387).(4)the expression level of YAP,CyclinD1 and MST1 was not related to the overall survival time of nasopharyngeal carcinoma patients after radiotherapy and chemotherapy.(5)There was no significant difference between the expression of YAP1 mRNA in 40 cases of nasopharyngeal carcinoma and 40 cases of nasopharyngeal mucosa chronic inflammation(F=1.508,p=0.223>0.05).Conclusions:(1)The expression of YAP and CyclinD1 in NPC was obviously up-regulated,and was closely related to the pathological type of NPC;the expression of MST1 in nasopharyngeal carcinoma was obviously down regulated.The expression level of YAP was positively correlated with Cyclin D1,It is suggested that YAP may be related to the occurrence and development of nasopharyngeal carcinoma.(2)There was no significance between the expression of YAP1 mRNA in nasopharyngeal carcinoma and chronic inflammation of nasopharynx mucosa.(3)The changes of YAP,CyclinD1 and MST1 expression levels in NPC tissues,play an important role in the occurrence and development of the tumor.
