Obejective Mammalian sterile line 20-like kinase 1(MST1),Yes-associated protein(YAP)and Transcriptional coactivator with PDZ-binding motif(TAZ)are the main members of the Hippo signaling pathway,may play an important role in the pathogenesis of autoimmune diseases such as rheumatoid arthritis(RA).This study aimed to analyze the expression level of MST1 in the serum of RA patients,and to explore the clinical significance and diagnostic value of serum MST1 levels in RA patients.Methods 34 patients with RA,20 patients with SLE and 20 healthy persons were enrolled in this study.Serum levels of MST1,YAP and TAZ were measured by Enzyme-Linked ImmunoSorbent Assay(ELISA).Meanwhile,clinical indicators of those patients were collected.The levels of serum MST1,YAP,TAZ expression levels were compared between RA patients and control group.The correlation between MST1 and bone erosion,disease activity score(DAS28),erythrocyte sedimentation rate(ESR),C-reactive protein(CRP),anti-cyclic citrullinated peptide antibody(anti-CCP antibody),rheumatoid factor(RF-IgA,RF-IgM,RF-IgG)was analyzed.Results 1.The serum MST1 level in the RA group was significantly lower than that in the SLE group [(5164.98±3547.23)pg/ml VS(9790.5018±2445.19)pg/ml,P<0.001]),and notably decreased than that in the healthy control group[(5164.98±3547.23)pg/ml VS(10238.99±1658.33)pg/ml,P<0.001].There was no statistical difference between the SLE group and the healthy control group([(9790.5018±2445.19)pg/ml)VS(10238.99±1658.33)pg/ml,P=0.508]).2.There was no significant difference in serum YAP level between RA patients and SLE patients([(0.1242±0.07292)ng/ml VS(0.1368±0.0885)ng/ml),P=0.459>0.05]),and no significant difference between RA patients and healthy controls([(0.1242±0.07292)ng/ml VS(0.1238±0.0635)ng/ml),P=0.987>0.05]).3.There wasno significant difference in serum TAZ level between RA patients and SLE patients([(0.5218±0.05424)ng/ml VS(0.5038±0.03887)ng/ml),P=0.266>0.05]),and no significant difference between RA patients and healthy controls[(0.5218±0.05424)ng/ml VS(0.4972±0.00709)ng/ml P=0.059>0.05].4.Serum MST1 level was negatively correlated with bone erosion(r=-0.5396,P=0.001<0.01),DAS28 score(r=-0.3773,P=0.0278<0.05),ESR(P=0.42>0.04),CRP `(P=0.5529>0.05),anti-ccp antibody(P=0.7321>0.05),RF-IgA(P=0.6680>0.05),RF-IgM(P=0.8639>0.05),and RF-IgG(P=0.9386>0.05)in RA patients.5.The area under ROC curve of serum MST1 level for diagnosis of RA is0.860 [95% CI = 0.757-0.964].When MST1 concentration is 1028 pg/ml,the sensitivity and specificity for diagnosis of RA are 60% and 88.2%.Conclusion 1.Compared with the control group,the level of serum MST1 in patients with RA was significantly lower,indicating that the expression of MST1 was related to the pathogenesis of RA.2.The level of serum MST1 in RA patients was negatively correlated with the degree of bone erosion,and negatively correlated with the DAS28 score,indicating that MST1 participated in the occurrence and development of bone destruction in RA and was related to the disease activity of RA.3.The serum MST1 level may be used as a new marker for the diagnosis of RA.
