Preliminary Study On Expression And Function Of CMTM7 Gene In Hepatocellular Carcinoma

Objective: to determine the expression of CMTM7 in hepatocellular carcinoma(HCC)and para cancer tissues,and to analyze the relationship between the expression of CMTM7 and the clinicopathological features and prognosis of HCC patients,and to detect the effect of CMTM7 on the biological function of hepatoma cells in vitro.Methods: the expression of CMTM7 in cancer and adjacent tissues of 75 HCC patients was detected by immunohistochemical technique,and the relationship between the expression of CMTM7 and the clinicopathological features and prognosis of the patients was analyzed.QRT-PCR and Western blot were used to detect the expression of CMTM7 at the level of mRNA and protein in the cancer and Para cancerous tissues of 20 HCC patients.Transfection of Sk-hep-1 and Bel-7402 cell lines with overexpressed plasmids,qRT-PCR and Western blot to verify the transfection efficiency;and Sk-hep-1 and Bel-7402 cells transfected with CMTM7 overexpressed plasmids were used to detect and observe the changes of cell biological function(such as CCK-8,cell clone formation,scratch test,Transwell experiment)And so on).Results:(1)the results of immunohistochemistry,qRT-PCR and Western blot showed that the expression of CMTM7 in HCC was significantly lower than that of para cancerous tissue(P < 0.05).The expression of CMTM7 was related to TNM staging,serum AFP level and distant metastasis.The low expression of CMTM7 suggests that HCC patients have poor prognosis.(2)after transfection of Sk-hep-1 and Bel-7402 cells with CMTM7 overexpression plasmids,the proliferation,migration and invasion of hepatoma cells were significantly decreased(P < 0.05).Conclusion:(1)CMTM7 is poorly expressed in HCC and is associated with poor prognosis.(2)over expression of CMTM7 in Sk-hep-1 and Bel-7402 hepatoma cell lines can inhibit the proliferation,invasion and metastasis of hepatoma cells.
