PDLIM1 Expression In Gastric Cancer And Its Correlation Between Clinicopathology And Prognosis

Abstract/Summary:
Objective:To investigate the expression of PDLIM1 in gastric cancer tissues,pericarcinomatous tissues and gastric cancer cell lines,and to explore the relationship between the expression level and the clinicopathological characteristics of gastric cancer patients.To investigate the effect of PDLIM1 expression on the prognosis of gastric cancer patients.Methods:134 specimens of gastric cancer tissues and pericarcinomatous tissues were collected from the First People’s Hospital of Changzhou between 2010 to 2011.All patients involved in the study were diagnosed with gastric cancer for the first time,and had never received any neoadjuvant chemotherapy,and were not associated with other types of malignancies.The patient had no family history of hereditary malignancy and all medical records were complete and accurate.At the same time,20 pairs of fresh gastric cancer tissues and pericarcinomatous tissues were collected,and three gastric cancer cell lines SGC-7901,BGC-823,MGC-803 and normal gastric mucosal cell line GES-1were cultured.(1)We used the Immunohistochemical method to detect the expression of PDLIM1 in gastric carcinoma and its pericarcinomatous tissues.(2)The expression of PDLIM1 m RNA in fresh gastric cancer and its pericarcinomatous tissues was detected by quantitative real time polymerase chain reaction(q RT-PCR).(3)q RT-PCR and Western blot were used to detect the expression of PDLIM1 m RNA and protein in three gastric cancer cell lines and the normal gastric mucosal cell line.(4)The relationship between PDLIM1 protein and clinicopathological features was examined by SPSS 19.0 statistical software package.Survival analysis was performed by Kaplan-Meier method,and the effect of PDLIM1 protein expression on the prognosis of gastric cancer patients was analyzed by Log-rank test.Results:(1)Among 134 gastric cancer tissues,56 cases(41.8%)showed high expression of PDLIM1 protein,and 70 cases(52.5%)showed high expression of PDLIM1 protein in paracancerous tissues.There was no obvious statistic difference.(2)In q RT-PCR experiment,the level of PDLIM1 m RNA in gastric cancer tissues was lower than that in pericarcinomatous tissues.The expression of PDLIM1 m RNA in three gastric cancer cell lines and corresponding normal gastric mucosa cell lines was lower than that in normal gastric cancer cell lines,and the SGC-7901 expression level was the lowest.(3)After Western blot experiment,the expression level of PDLIM1 protein in three gastric cancer cell lines were lower than that in normal gastric mucosa cell lines,and the SGC-7901 expression level was the lowest.(4)The expression of PDLIM1 protein was significantly correlated with the low expression of PDLIM1 protein in tumor size(P=0.019),vessel invasion(P=0.005),distant metastasis(P=0.021),lymph node metastasis(P=0.008)and TNM stage(P=0.001).(5)The 5-year survival rate of gastric cancer patients with low expression of PDLIM1 was significantly lower than that of patients with high expression of PDLIM1(P=0.029).The median cumulative survival time of patients(27 months;95% CI: 27.552-35.896 months)with low expression of PDLIM1 was significantly lower than that of high expression patients(39months;95% CI,35.115-44.814 months).Conclusion:(1)The expression of PDLIM1 in gastric cancer was lower than that in normal tissues.The results suggest that PDLIM1 may play an important role as a tumor suppressor gene in gastric cancer.(2)The expression of PDLIM1 in gastric cancer is significantly different from that of lymph node metastasis,distant metastasis and TNM staging in gastric cancer.The results suggested that the expression level of PDLIM1 protein might be related to the invasion and metastasis ability of gastric cancer cells.(3)The total survival time of patients with positive expression of PDLIM1 protein was significantly prolonged,suggesting that PDLIM1 protein might be an independent factor in evaluating the prognosis of gastric cancer patients.
