Objectives: To Observe the effects of vitamin D intervention on ovarian function in rats with polycystic ovary syndrome.Methods: Twenty-four 6-week-old female rats were divided into the control group,the PCOS group and the PCOS+VD group.In the PCOS group,the rats were given intragastric administration of letrozole 1mg/Kg/d in the morning and 0.8ml/Kg/d of peanut oil in the afternoon.The PCOS+VD group was treated with letrozole 1mg/Kg/d in the morning,and calcitriol 0.2ug/Kg/d in the afternoon.The control group was given a corresponding dose of carboxymethyl cellulose solution and peanut oil per day.Vaginal smears were examined to monitor changes of rats oestrous cycle,and the rats' weights were recorded daily.At the end of the 28 days,blood samples were collected and the ovarian tissues were taken for histological examinations.Serum testosterone(T),luteinizing hormone(LH),follicle stimulating hormone(FSH),estradiol(E2),progesterone(P),Anti-Mullerian hormone(AMH),and Advanced glycation end products(AGEs)were measured;histologic changes in ovaries were observed by HE staining.Results: Compared with the control group,the serum 25(OH)D level in the PCOS group was significantly lower,serum T,LH,AMH,AGEs levels were significantly higher,(P<0.05),and the oestrous cycle of the PCOS group rats were disturbed,ovarian HE staining shows a large number of cystic follicles,and the number of corpus luteum were decreased.Compared with the PCOS group,the serum 25(OH)D level in the PCOS+VD group was increased,and serum T,LH,AMH and AGEs were decreased(P>0.05);the oestrous cycle was relatively regular,the number of characteristic cystic follicles was reduced,and the number of corpus luteum was Increased in the PCOS+VD group.Conclusions:(1).Our results show that letrozole-induced PCOS rat model is successful.(2)Compared with PCOS group,serum T,LH,AMH and AGEs were decreased in the PCOS+VD group.The oestrous cycle and histological appearance of ovarian sections were also improved,which partially restored the ovarian function of PCOS rats.
