Studies On The Chemical Compasitions Of The Ethanol-extraction Of A.sisalana Perr.

Guangxi provides abundant A.sisalana Perr.resources,but a lot of the residue has been left brushed off during industrial processing.In order to improve the utilization rate of A.sisalana Perr.,to guide the industrial production,and to reduce the pollution of the environment,it’s very necessary to analyse the chemical composition of the residue from A.sisalana Perr.So more value of A.sisalana Perr.will present.Former research showed that the extract of A.sisalana Perr.can be treated as a kind of chinese medicine to detoxify,discharge pus,and lower blood-lipid.It can be used in the treatment of,phthisis,apostaxis,hematochezia,dysentery,ulcers,hemorrhoids,and so on.The fresh leaves can be use to treat carbuncle.Therefore,the research of the chemical composition and their biological activity of the residue from A.sisalana Perr.,can make full use of A.sisalana Perr.resources.At present,e.g.,saponins(sapogenin),polysaccharides,amino acid are isolated from A.sisalana Perr..But there is not a systematic research carrying on low-polar petroleum ether fraction,ethyl acetate fraction,and water-extraction fraction of ethanol-extraction from A.sisalana Perr..This paper shows that,the washing dry A.sisalana Perr.mesophyll were heated to a boiling point in the ethanol to get ethanol-extraction.After the ethanol-extraction was concentrated,the low-polar petroleum ether fraction,ethyl acetate fraction,and water-extraction fraction,were respectively isolated from ethanol-extraction by using petroleum ether,ethyl acetate,and water.Then these three fractions of extraction have been researched.The chemical composition of ethyl acetate fraction were extracted,isolated,and purified by using column chromatography method,combining with various spectrum testing(including IR,SPME/GC/MS,ESI-MS,NMR and X-ray)to get the structure identification.Four compounds were found,they all belong to saponins(sapogenin),including:gitogenin,hongguanggenin,and a new saponins,which is named as hecogenin 4-ethylene glycol glucose,and a compound structure was preliminaryly determined as a new structure of the association of two saponins.Among them,the single crystal data of the crystals of hongguanggenin-methanol were first reported(Cambridge Crystallographic Data Centre Number:CCDC 1267/35 73).The chemical composition of water-extraction fraction were extracted,isolated,and purified by using column chromatography method,combining with various spectrum testing(including IR,ESI-MS,NMR)to identify the structures.Two compounds were found,they all belong to saponins(sapogenin).The chemical composition of the low-polar petroleum ether fraction were preliminaryly determined by using SPME/GC/MS method.And the total ion chromatrogram(TIC)was set up by this testing.Then,the data of mass spectrometry was searching in the database of NIST,and the content of each constituents were quantified with the peak area normalization method.Seventeen kinds of constituents were preliminarily determined,including:2,4-di-tert-butyl-Phenol;1-Hexadecene;1-Octadecene;6,10,14-Trimethyl-2-pentadecanone;Butanoic acid,3,7-dimethyl-6-octenyl ester;1,2-Benzenedicarboxylic acid,diheptyl ester;Hexadecanoic acid,methyl ester;n-Hexadecanoic acid;Hexadecanoic acid,ethyl ester;9,12-Octadecadienoic acid(Z,Z)-,methyl ester;Linolenic acid,methyl ester;3,7,11,15-Tetramethyl-2-hexadecen-1-ol;9,12-Octadecadienoic acid(Z,Z)-;2-Methyl-Z,Z-3,13-octadecadienol;9,12-Octadecadienoic acid,ethyl ester;1,2-Benzenedicarboxylic acid,diisooctyl ester;2,6-Di-tert-butyl-4-[(2-octadecyloxycarbonyl)eythyl]-phenol.
