Objective:The usage of Rhubarb has a long history which is one of the common traditional Chinese medicines.It was composed with different processed drugs to prescription which can realize its unique therapeutic action.The core of Chinese medicine processing is the change of property with the change of effective material basic.The research of 6 Rhubarb processed drugs were raw Rhubarb,steaming prepared rhubarb,prepared rhubarb,rhubarb root parched in wine,rhubarb root parched in vinegar and rhubarb charcoal.They were processed with Mangnolia officinalis,Immature bitter orange and Mirabilite.The purgative activity,endotoxin,NO and TNF-?were dected with pharmacology and efficacy experiments.HPLC was used to determine the effects of different Rhubarb processed drugs to active ingredient content change of Dachengqitang.The analytical methods of in“ingredient-efficacy”in Dachengqitang of different Rhubarb processed drugs were established.The active ingredients and efficacy impacts of different Rhubarb processed drugs to Dachengqitang prescription and its correlation were discussed.The research explained mechanism of the impact change of Chinese medicine processed drugs to active ingredients and efficacy prescription,and provided new thought and method to prescription of different Chinese medicine processed drugs.Methods:Referencing“Chinese traditional Chinese medicine yinpian processing standard”,“Chinese medicine processing”and“Chinese Pharmacopoeia”of 2015 edition,the different Rhubarb processed drugs were processed.The purgative activity,charcoal prograding,Na+-K+-ATPase activity of mice parietal cell,endotoxin in mice serum,NO and TNF-?were researched.And the content impacts of purgative activity,charcoal prograding,Na+-K+-ATPase activity of mice parietal cell,endotoxin in mice serum,NO and TNF-?with different Rhubarb processed drugs were compared.With HPLC,chromatographic was Agilent Zorbax SB-C18 5?m?4.6mm x 250mm?,and mobile phase was acetonitrile-0.1%phosphoric acid solution.Through gradient elution,flow velocity was 1ml·min-1,and detection wave length were280nm and 254nm.In 30?of column temperature,10 active ingredients in Dachengqitang of 6 different Rhubarb processed drugs were determined and the content variation was analyzed.Result:1.Through processing with formal processed methods,6Rhubarb processed drugs all met medical standard.2. Six Dachengqitang of different Rhubarb processed drugs had varying degrees impacts on purgative activity,but the variation differences had certain universal relationships with Rhubarb processed drugs.Raw Through comparison between DCQT group,steaming DCQT group,ripe DCQT group,wine DCQT group,vinegar DCQT group,charcoal DCQT group and blank control group,positive control group,the effects of experiment groups in purgative activity,charcoal prograding,Na+-K+-ATPase inhibition activity were different.Within d 6 experiment groups,Raw DCQT group had most significant difference.The 6 g·kg-1 group and 10g·kg-1 group all had differences.3. In all Dachengqitang experimental groups,except for charcoal DCQT group,all can softened excrement and promoted fecal peritonitis mice with hot choking to defecate.In water intake of fecal peritonitis mice,each Dachengqitang groups were higher than positive group and model group except for charcoal DCQT group.4. All different Rhubarb processed drugs Dachengqitang could increase the content of NO in ABP mice serum,but compared with other groups,the effects of NO content increasing in ABP mice serum was not significant in charcoal DCQT group.5. All different Rhubarb processed drugs Dachengqitang could decrease the content of TNF-?in ABP mice serum,but compared with other groups,the effects of TNF-?content reduction in ABP mice serum was not significant in charcoal DCQT group.6. Ten standard curves of 10 active ingredients in different Rhubard processed drugs Dachengqitang all had well linear relation,and the methodology inspects reached the experimental requirement.10 active ingredients of different Rhubarb processed drugs Dachengqitang had varying degrees of change.Compared with Raw DCQT group,anthraquinones in other groups all decreased.The charcoal DCQT group had maximum degree of reduction.The changes of other ingredients were not significant.Conclusion:The experiment aimed to research the impacts of different Rhubarb processed drugs Dachengqitang to purgative activity,endotoxin,NO and TNF-?,and the purgative activity,endotoxin,NO,TNF-?and 10 active ingredients content had varying degrees of changes.These changes were related to the compatible Dachengqitang of different Rhubarb processed dugs,and content change of Rhubarb anthraquinones had certain impacts and universal relationships on the efficacy of Dachengqitang.
