Objective:Post-infectious cough(PIC,Cough after a cold)is caused by upper respiratory tract infection,a class of coughs that remain unchanged after the infection being controlled.It belongs to a sub-acute cough as a common disease in the department of respiratory medicine.There has been no specific treatment so far.Clinically,symptomatic treatment is a major therapy.Antihistamines,central antitussive drugs and hormone are often selected,but it is effective only for some patients and has many side effects and it is easy to relapse after stopping administration.The Ganhouke is a prescription for the treatment of PIC,which is effective in clinical application.The extract used in this study is extracted from Ganhouke by modern separation techniques.In this study,animal models were used to evaluate the pharmacodynamics of Ganhouke extract(GHKE)for the treatment of PIC,and the possible mechanism was discussed.The results of this study provide experimental basis for the development and utilization of traditional Chinese medicine.Methods:1.The pharmacodynamics study of GHKE on animal models1.1 Effects on antitussive of GHKECough was induced by ammonia in mice,the latent period and the number of cough in 3 minutes were observed.Cough was induced by the inhalation of 17.5%citric acid in guinea pigs,the latent period and the number of cough in 6 minutes were observed.1.2 Effects on asthma of GHKE in guinea pigsAsthma was induced by breathing in acetylcholine chloride-histamine phosphate in guinea pigs,the latent period of asthma and falling animal number were observed.1.3 Anti-inflammatory effect of GHKE in miceThe ear swelling was observed using xylene-induced mouse ear edema model.1.4 Dispelling phlegm effect of GHKE in miceThe excretion of phenol red was observed in phenolsulfonphthalein excretion model in mice.2.The effect of GHKE on the contraction of bronchial smooth muscle of guinea pigs in vitroThe guinea pig's trachea was isolated and tracheal ring was made in vitro.The contractile function of tracheal ring induced by histamine was measured.Results:1.The pharmacodynamics study of GHKE on animal models1.1 Effects on antitussive of GHKEGHKE could significantly prolong the latent period of cough induced by ammonia in mice with the dosage of 0.4g/kg and 0.8g/kg and decrease the number of cough in 3 minutes induced by ammonia in mice with the dosage of 0.2g/kg and 0.4g/kg after the administration for 7 days compared with the contral group.GHKE could significantly prolong the latent period of cough and decrease the number of cough induced by citric acid in guinea pigs with the dosage of 0.1g/kg,0.2g/kg and 0.4g/kg after the administration for 7 days,compared with the contral group.1.2 Effects on asthma of GHKE in guinea pigsGHKE could significantly prolong the latent period of asthma induced by breathing in acetylcholine chloride-histamine phosphate in guinea pigs and decrease the falling animal number compared with the contral group with the dosage of 0.2g/kg and 0.4g/kg after the administration for 7 days.1.3 Anti-inflammatory effect of GHKE in miceGHKE could significantly decrease the ear swelling induced by xylene-induced in mice compared with the contral group with the dosage of 0.4g/kg and 0.8g/kg after the administration for 7 days.1.4 Dispelling phlegm effect of GHKE in miceGHKE could significantly increase the phenolsulfonphthalein excretion in mice compared with the contral group with the dosage of 0.8g/kg after the administration for 7 days.2.The effect of GHKE on the contraction of bronchial smooth muscle of guinea pigs in vitroGHKE at the concentration of 20,40mg/ml could inhibit the contraction of isolated trachea smooth muscle of guinea pigs induced by hitamine in vitro.Conclusion:GHKE possesses significantly antitussive effect in mice and guinea pigs,alleviates asthma induced by breathing in acetylcholine chloride-histamine phosphate in guinea pigs,inhibit the contraction of isolated trachea smooth muscle of guinea pigs induced by hitamine in vitro,decreases the ear swelling induced by xylene-induced in mice and increases the phenolsulfonphthalein excretion in mice.The results showed that GHKE possess antitussive,antiasthmatic and anti-inflammatory effects,dilate the bronchus and relieve the symptoms of respiratory tract.
