Objective: To study the changes of GAP-43 protein in peripheral blood of patients with first-episode schizophrenia and its relationship with psychotic symptoms.Methods: The patient group included 18 first-episode schizophrenia patients aged 18 to 35 years old who were hospitalized in the No.7 People's Hospital of Dalian from 2016 to 2017 and were given risperidone orally disintegrating tablets(4-8 mg/day)to improve psychotic symptoms.The control group randomly selected 18 to 35-year-old staff of the hospital.For the patient group,PANSS,Wisconsin card sorting tests and GAP-43 levels in the serum have been tested at the time of admission and after oral administration of risperidone for 6 weeks.Only measure serum GAP-43 content in the control group once.The serum level of GAP-43 protein was compared between the patient group and the control group,and the level of serum GAP-43 protein before and after the intervention of the patient group was compared.The decrease of GAP-43 content and the decrease ratio of the serum of the patients after intervention and the total score reduction rate of the PANSS scale,the rate of subtraction and subtraction of the positive symptoms,the correlation of the negative symptom subtraction rate and the general pathological symptom reduction rate were compared.The serum GAP-43 content and the total score of PANSS,positive symptoms,negative symptoms and general pathological symptoms were analyzed before and after intervention.The correlation between the level of GAP-43 protein before and after intervention was analyzed with the scores of RA,CC,RC,RE,RF,RP,RPE,NRPE,and WSCT.Results: The level of GAP-43 protein in peripheral blood before intervention was significantly higher than that in the control group,the difference was statistically significant(t = 10.672,P <0.001).After 6 weeks of risperidone intervention,the GAP-43 level of the patients was still significantly higher than that of the control group,the difference was statistically significant(t = 3.487,P < 0.01).After intervention,the level of GAP-43 in patients was significantly decreased,and the difference was statistically significant(t =8.042,p < 0.001)(GAP-43 content: before intervention:373.46 + 50.36,after intervention:293.41 + 63.11,the control group:239.67 + 16.65).PANSS total score,positive symptom score(P),negative symptom score(N),and general pathological symptom score.(G)decreased significantly(t = 33.600,29.817,22.179,34.620,P < 0.01),and RP in Wisconsin card evaluation results,RPE scores decreased,and the difference was statistically significant(t = 2.535,2.411,P < 0.05),RA,CC,RC,RE,RF,NRPE,FM have no difference(t = 1.099,0.119,-0.810,1.426,-0.383,-0.535,p > 0.05).After the intervention,the decrease and decrease ratio of serum GAP-43 and the total score reduction rate of PANSS,the P subtraction rate,the N subdivision rate and the G score reduction rate were not related(P > 0.05).Before the intervention,the expression of GAP-43 was not related to the total score of PANSS,P,N,and G(P > 0.05),the total subtraction of GAP-43 and PANSS,the P subtraction,N subtraction,G subtraction(P > 0.05).The scores were not related(P > 0.05);the total scores of GAP-43 and PANSS,the P subtraction,the N subtraction,and the G subtraction were not related(P > 0.05),and GAP-43 and WSCT(RA,CC,RC,RE,RF,RP,RPE,NRPE,FM)were not related(P > 0.05).Conclusion: 1.GAP-43 protein is associated with the onset of schizophrenia;2.The level of serum GAP-43 in schizophrenic patients was influenced by the state of illness and risperidone intervention.;3.The increase of GAP-43 in the episode of schizophrenia may be an increase in the reactivity caused by the reduction of synapse density caused by the disease.
