Objective To investigate the effect of Electroacupuncture(EAS)of "Zusanli" on pain behavior,inflammatory factors and related proteins in formalin acute inflammatory pain rats.Methods 32 male SD rats,2~3 months old,weight 250-300 g,were selected as the research object.According to randomized principle,all rats were divided into a control group,model group,electroacupuncture pre-stimulation group and electroacupuncture prostimulation group with 8 rats in each group.The control group: According to the operation requirements of electroacupuncture,rats were bundled(30minutes / time / day ×2 days).During this period,the rats were not treated with electroacupuncture and 50 ul 0.9% saline was injected to the right foot of the rats at the last binding;The model group: According to the operation requirements of electroacupuncture rats were bundled(30 minutes / time / day×2 days).During this period,the rats were not treated with electroacupuncture and 50 ul 5% formalin was injected to the right foot of the rats at the last binding to construct acute inflammatory pain model rats.The electroacupuncture pre-stimulation group: According to the operation requirements of electroacupuncture rats were bundled and electroacupu-ncture stimulation was given(30 minutes / time / day ×2 days),50 ul 5% formalin wasinjected to the right foot of the rats at the last binding;The electroacupuncture pro-stimulation group: According to the operation requirements of electroacupuncture rats were bundled,and 50 ul 5% formalin was injected to the right foot of the rats at the last binding,and then the electroacupuncture stimulation was given(30 minutes / time ×2 times).At the end of the above operation,the spontaneous pain behavior of rats was observed within 1 hour:the number of 1H licking feet,the times of 10 min contraction,and the time of10 min licking.At the end of the two hour time points,each group were randomly selected 4 rats for enzyme-linked immunosorbent assay(ELISA)test of proinflammatory cytokine tumor necrosis factor-?(TNF-?),interleukin-1?(IL-1?)and interleukin-6(IL-6),and anti-inflammatory factor interleukin-10(IL-10).The remaining 4 rats of the each group were taken to prepared the lumbar enlargement of the spinal cord sections for immunohistochemical staining test of spinal dorsal horn Iba1 positive microglia,substance P(SP)positive terminals and the expression level of FOS protein.Results When compared with the control group,the 1H licking feet,the times of 10 min contraction,and the time of 10 min licking of the model group increased(P<0.05);the TNF-?,IL-1?,IL-6 increased(P<0.05);the IL-10 decreased(P<0.05);while the spinal dorsal horn Iba1 positive microglia,SP and the IOD/S of the FOS protein increased(P<0.05).When compared with the model group,the 1H licking feet,the times of 10 min contraction,and the time of10 min licking of the electroacupuncture pre-stimulation group or theelectroacupuncture pro-stimulation group increased(P<0.05);the TNF-?,IL-1?,IL-6 increased(P<0.05);the IL-10 decreased(P<0.05);while the spinal dorsal horn Iba1 positive microglia,SP and the IOD/S of the FOS protein increased(P<0.05).When compared with the electroacupuncture prostimulation group,the 1H licking feet,the times of 10 min contraction,and the time of 10 min licking of electroacupuncture pre-stimulation group increased(P<0.05);the TNF-?,IL-1?,IL-6 increased(P<0.05);the IL-10 decreased(P<0.05);while the spinal dorsal horn Iba1 positive microglia,SP and the IOD/S of the FOS protein increased(P<0.05).Conclusions ?The electroacupu-ncture stimulation of Zusanli can relieve the acute inflammatory pain rats,electroacupuncture pre-stimulation got better effects.? The electroacupuncture stimulation of Zusanli can decreased the proinflammatory cytokine factors and increased the anti-inflammation cytokines,but the electroacupuncture pre-stimulation got better effects.? The electroacupuncture stimulation of Zusanli can decreased the spinal dorsal horn Iba1 positive microglia,SP and the IOD/S of the FOS protein,but the electroacupuncture pre-stimulation got better effects.
