Experimental Study On The Effect Of Electroacupuncture At "Zusanli" And "Quchi" On Inflammatory Factors In Prehypertensive Rats

Purpose:Observing interleukin-6, interleukin-1β, interferon-γ content and renal tumor necrosis factor-α m RNA expression in prehypertension in rats serum by Electro-acupuncture of “Zusanli”and“Quchi”point, to explore whether abnormal inflammatory factor is one of the pathological factors of prehypertension, and to analyze and discuss curative effect and mechanism of prehypertension electro-acupuncture treatment. Materials and methods:30 healthy male Dahl salt sensitive rats(DS), were randomly divided into 3 groups, 10 rats in each group, namely, model group, acupuncture group, and non acupoint group; normal group for 10 healthy male salt resistant rats(SS). After successfully modeling, 4 groups were feed normally; model group daily bundled and fixed for 20 min without acupuncture; Rats in the model group fixed 20 min daily without acupuncture;acupuncture group selected bilateral “Quchi” “Zusanli”acupoints in rats according to “experimental acupuncture science”,and inserting by using filiform needle about 3mm for electro-acupuncture(EA parameters:density wave,frequency 2Hz) intensity was accorded with trembling of needle handle,and stimulation for 20 min,once a day,6 times per week for treatment,and continuously for 4 weeks;The selection of“Zusanli”acupoint in rats for non acupoint group was corresponding points beside the “Zusanli” 1-2mm,corresponding points for“Quchi” was selected in junction of upper arm front deltoid and biceps, and treatment methods and time were same with acupuncture group.Take out serum, kidney tissue from the rats after the end of treatment.Use ELISA to detect content in rat serum interleukin-6 levels, interleukin-1βand interferon-γ; use fluorescence quantitative PCR method to detect renal tumor necrosis factor alpha m RNA expression. Results: 1.Effects on each rat tail artery systolic pressure by electro-acupuncture stimulating “Zusanli”and“Quchi” acupointCompared with normal group rats, rat tail artery systolic pressure in the model group significantly increased(P<0.01).Compared with model group rats,rat tail artery systolic pressure in the acupuncture group and non acupoints group significantly decreased(P<0.01).The acupuncture group was compared with the normal group,which was not statistically significant(P>0.05). 2.Effects for interleukin-6 content on prehypertension rat by electro-acupuncture at bilatera “Zusanli”and“Quchi” acupointCompared with the normal group rats, IL-6 content in serum of rats in model group increased significantly(P<0.01); after 28 days EA treatment, compared with model group, IL-6 content in acupuncture group obviously decreased(P<0.01); the non acupoint group was compared with model group, IL-6 content was also lower(P<0.01); IL-6 content in acupuncture group was significantly lower than normal group(P<0.01). 3.Effects for interleukin-1β content on prehypertension rat by electro-acupuncture at bilatera “Zusanli”and“Quchi” acupointCompared with the normal group rats, IL-1β content in serum of the rats in the model group increased significantly(P<0.01); after 4 weeks EA treatment, compared with model group, IL-1β content in acupuncture group obviously decreased(P<0.01);IL-1β content in the non acupoint group was compared with the model group which was lower(P<0.01); the acupuncture group was obviously lower than that of normal group(P<0.01). 4.Effects for interferon-γ content on prehypertension rat by electro-acupuncture at bilatera “Zusanli”and“Quchi” acupointCompared with the normal group rats, IFN-γ Content in serum of the rats in the model group increased significantly(P<0.01); after 4 weeks EA treatment, compared with model group, acupuncture group of IFN-γ content decreased(P<0.01); acupuncture group of IFN-γ content was significantly lower than the normal group(P<0.01);IFN-γ content in the non acupoint group was compared with model group,which was no statistical significance(P>0.05).5.Electro-acupuncture at bilateral “Zusanli”and“Quchi” acupoint can improve the prehypertension rat kidney tumor necrosis factor alpha gene expressionCompared with the normal group rats, the expression quantity of renal TNF-α gene in model group rats increased significantly,(P<0.01); after 4 weeks EA treatment, compared with model group, TNF-α gene expression in acupuncture group were significantly decreased(P<0.01); TNF-α gene expression content in the non acupoint group was compared with that of model group,which was no statistical significance(P> 0.05). Compared with the acupuncture group,the normal group in TNF-α gene expression content was no significant difference(P>0.05). Conclusion: 1. Electro-acupuncture “Zusanli” and “Quchi” acupoint can effectively reduce the prehypertension systolic arterial pressure. 2. The level of inflammatory factors for prehypertension rats is abnormal, which may be one of the pathological mechanism of prehypertension. 3. Electro acupuncture“Zusanli” and “Quchi” point can decrease the prehypertension systolic arterial pressure by regulating inflammatory factor expression.
