Objective: To study region-specific membrane proteins in Cochlear nucleus(CN)and analyze functions and their role in central auditory transmission.Methods: The CN region-specific membrane proteins on P60 male SD rats(weight:230g)were seperated with iTRAQ(isobaric tags for relative and absolute quantitation).The Rest of the brain was used as the control.The mass spectrometry was used for qualitative and quantitative analysis of protein spots.Then the specific membrane protenins were identified.By Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis,the potential functions of region-specific proteins were proposed,and the specific proteins related to auditory processing disorder(APD)were identified.Results: Totally 17 membrane proteins of CN were detected.GO pathway hint they mainly take part in the regulation of multicellular organism process and development.Among them,there were 7 genes involved in biological regulation;6 genes involved in the growth and development;5 genes involved in cellular component assembly and celluar development process;4 genes involved in response to inorganic substance and wounding,protein complex biogenesis,homeostatic process,cell death,macromolecular complex subunit organization and celluar response to chemical stimulus;3 genes involved in coagulation,regulation of body fluid levels and muscle structure develpoment.KEGG pathway analysis find 4 significant enrichment pathways(amyotrophic lateral sclerosis(ALS),complement and coagulation cascades,Influenza A,neuroactive ligand-receptor interaction).Peripherin(PRPH)and neurofilament proteins involved in ALS and these proteins aggregation can cause axonal transport disorder,which leads to neurodegenerative disease.Uniprot showed Galectin-1 was associated with nerve growth and development.PRPH is involved in amyotrophic lateral sclerosis(ALS)process related to neurodegenerative disease.Annexin A2 participates in a variety of biological regulation processes.Galectin-1 plays an important role in the development of central nervous system,axon regeneration and synaptic plasticity.Peripherin can cause neurotoxicity when it expressed highly.Peripherin leads to ALS together with neurofilament proteins.Annexin A2 has a neurotrophic effect,and may regulate exocytosis of synaptic vesicle at the nerve terminals.Conclusion: Galectin-1,Annexin A2 and Peripherin were CN-specific membrane proteins,which might play some important roles in neural development,synaptic plasticity and neural degenerative diseases.The results provided theoretical basis for further research on the pathogenesis of CAPD.
