ObjectiveIn this study,various indicators and related endogenous substance levels in chronic stress model rats were observed through behavioral,pathological,molecular,and plasma metabonomics studies to assess chronic chronic stress.Effect of Heterotypic Oxidative Stress(HIS)Model on Rats and Intervention Effect of Electroacupuncture.At the same time,by observing the structure and morphology of adrenocortical cells in different groups of rats,the mechanism of electroacupuncture on the protective effect of glycometabolism in chronic stress model rats was explained from the perspective of adjusting HPA axis.MethodsSD rats were randomly divided into normal group,HIS model group(abbreviated as model group)and model electroacupuncture group(referred to as electroacupuncture group).The model group and the electroacupuncture group randomly used three different stimulation methods once a day for 9 days to create the HIS model.The changes of body weight and thermal radiation latency before and after modeling were compared.At 9 am to 11 pm on the second day after the completion of the modeling,the rats were killed by decapitation and the plasma,hypothalamus,and adrenal glands of the rats were collected.The structure and morphology of adrenocortical cells in rats were observed using a smart biological microscope.Cortisol(CORT)and corticotrophin-releasing hormone(CRH)levels in the hypothalamus,adrenal gland,and plasma were measured by enzyme-linked immunosorbent assay(ELISA).Variety.Using nuclear magnetic resonance spectroscopy 1H-NMR metabolomics technology to analyze the changes of endogenous metabolites in plasma,using multivariate statistical methods to identify the change of plasma metabolic profiles in each group of rats,screening plasma biomarkers.Results(1)Behavioral results showed that:1 Compared with the normal group,the body mass gain of the model group was significantly lower than that of the normal group(p<0.01);compared with the model group,the body mass of the electroacupuncture group increased significantly more than the model group(p<0.01).After rats were modeled with thermal pain threshold,compared with the normal group,the thermal radiation latency of the model group was significantly decreased(p<0.05).Compared with the model group,the thermal radiation latency of the electroacupuncture group was significantly increased(p<0.05).(2)Histopathological section of adrenal gland showed:Compared with the normal group,the adrenal glomus of the model group became thinner,irregularly arranged,and the nucleus was significantly reduced.Compared with the model group,electroacupuncture can reduce the changes of morphology and tissue structure of adrenocortical cells in HIS rat models.(3)The ELISA results of plasma in each group showed:1 Comparison of plasma samples,compared with normal group,decreased expression of corticotropin releasing hormone(CRH)in plasma of model group(p<0.05),plasma cortisol(CORT)The content increased(p<0.05)and the difference was statistically significant.Compared with the model group,the expression of corticotropin-releasing hormone(CRH)in the plasma of the electroacupuncture group was increased(p<0.05),and the plasma cortisol(CORT)content was decreased(p<0.01).2 Comparison of adrenal samples:Compared with the normal group,cortisol(CORT)content in the adrenal glands of the model group decreased(p<0.05).Compared with the model group,cortisol(CORT)levels in the adrenal glands of the electroacupuncture group increased(p<0.05).).3 Comparison of hypothalamic samples:Compared with the normal group,the expression of corticotrophin-releasing hormone(CRH)in the model group was decreased(p<0.01);compared with the model group,the expression of corticotrophin-releasing hormone(CRH)was increased in the EA group.High(p<0.01).(4)1H NMR analysis showed that the levels of alanineandproline in the plasma of the model group were significantly higher than those in the control group(p<0.05),and glucose levels were significantly lower(p<0.05)..Compared with the model group,plasma alanine and proline levels in the electroacupuncture group were significantly lower(p<0.05),and glucose levels were significantly higher(p<0.05).ConclusionElectroacupuncture intervention can significantly reverse the state of glucose metabolism disorder in chronic stress model rats,and its protective effect is very obvious,which is closely related to the adjustment effect of electroacupuncture on HPA axis.
