Objective:To investigate the effect of asiaticoside(AS)on the blood lipids and lipid metabolism in the high-fat diet induced golden hamsters hyperlipidemia model,and provide theoretical supporting for clinical treatment for hyperlipidemia.Methods:First ly,the lipid-lowering activity of asiaticoside was screened by KM mouse model of acute hyperlipidemia,and then high-fat diet induced golden hamster model of hyperlipidemia was used to explore the effect of different concentrations of Centella asiatica glycosides on blood lipids and liver protection.1.Screening for acute hypolipidemic activity:KM mice were randomly divided into 4 groups:normal group,model group,fenofibrate group,asiaticoside group,10 mice in each group.An animal model of acute hyperlipidemia was established by intraperitoneal injection of Triton WR-1339(400 mg/kg)in model group,fenofibrate group and asiaticosid group.Fenofibrate and asiatica groups were gavaged with 100 mg/kg does of fenofibrate and asiatica glycoside solution respectively.The normal and model group were given the same amount of normal saline.After 16 hours,blood samples were taken to determine the total cholesterol(TC)and triglyceride(TG)levels in the serum of each mouse.2 high fat diet induced hyperlipidemia model:Syrian golden hamsters were randomly divided into 6 groups,namely the normal control group,hyperlipidemia model group,Xuezhikang group(250mg/kg),asiaticoside low dose Group(15 mg/kg),asiaticoside mid dose group(30 mg/kg),asiaticoside high dose group(60 mg/kg).In addition to the normal control group was given the basic feed,other experimental groups were given a high-fat diet to establish an animal model of hyperlipidemia.At the same time,the normal control group,hyperlipidemia model group were fed the same amount of saline,the other groups were administered the drugs according to the selected dose.The levels of TC,TG,low-density lipoprotein-cholesterol(LDL-C),high-density lipoprotein-cholesterol(HDL-C),and low-density lipoprotein(HDL-C)in serum were measured after 4 weeks adminstration.The levels of glutamate-private transaminase(ALT or GPT)and glutamic-oxalacetic transaminase(GOT or AST)in serum,as well as the levels of TC,TG,superoxide dismutase(SOD)and glutathione Peptide peroxidase(GSH-PX)in liver tissue were measured after 4 weeks treatment.Additionally,the structural changes of liver tissue were observed by hematoxylin-eosin staining(H&E Stainning).Results:1.The results showed that the serum level of TC and TG were decreased by asiaticoside in acute hyperlipidmia mice,which indicat that asiaticoside could regulate the concentrate of lipid in serum.2.The results showed that compared with the model group,the serum level of TC,TGS LDL-C,TC/HDL and AST,as well as the levels of TC and TG in liver were decreased by different does of asiaticoside in high-fat induced hyperlipidmia hamster.On the contrary,the levels of hepatic SOD and GSH-PX increased in asiaticoside group.The HE staining results illustrated that fat deposition in the liver was improved by administration of asiaticoside.Conclusion:Asiaticoside can effectively regulate blood lipids and significantly inhibit hepatic fatty lesions in hyperlipidemia golden hamsters through liver antioxidation.
