Expression And Prognostic Analysis Of Twist And N-Cadherin In Non-Small Cell Lung Adenocarcinoma

Abstract/Summary:
Objective: To investigate the differences in mRNA and protein expression levels of transcription factors Twist and N-cadherin in non-small-cell carcinoma(NSCLC)and the relationship between them.The relationship between the level of its mRNA and protein expression in tissues and the clinicopathological and prognosis of patients with non-small cell lung adenocarcinoma.Methods: 41 non-small cell lung adenocarcinoma tissues and their corresponding normal adjacent tissues and 12 benign lesions(January 2015 to May 2015)were collected to extract non-small cell lung adenocarcinoma and paracancerous tissues and benign lesions.The RNA in the RNA was then reverse transcribed into cDNA.The expression of Twist and N-cadherin in the RNA of each group was detected by qRT-PCR.The relationship between the expression of the RNA and the clinicopathologic features and the mRNA expression of the two genes were explored.The correlation;at the same time,the use of immunohistochemical SP method to detect the same specimens of non-small cell lung adenocarcinoma tissue and adjacent tissue and benign lesions in protein expression,and explore the expression differences,the same analysis of itsexpression and clinical The relationship between the pathology and both.Finally,combined with the results of these two factors and follow-up results to make a prognostic analysis.Results: Real-Time PCR showed that the expression of Twist mRNA in the observation group was significantly greater than that in the para-cancer group and the control group(P<0.01).The expression of N-cadherin mRNA in the observation group was significantly greater than that in the para-cancer group.And the control group,the difference was statistically significant(P <0.01).At the same time,the expression of Twist and N-cadherin mRNA in the non-small cell lung adenocarcinoma tissue was not significantly related to the patient’s gender,age,and differentiation(P>0.05).However,the expression of Twist and N-cadherin mRNA increased with lymph node Metastasis was related to TNM staging;in addition,Twist mRNA and N-cadherin mRNA were positively correlated at the RNA level(r=+0.563,P=0.000).In addition,in order to study the consistency of mRNA and protein expression in the same tissue,the Twist and N-cadherin protein levels of the same non-small cell lung adenocarcinoma specimens and benign lesions were detected by immunohistochemical SP method.The expression levels of Twist and N-cadherin proteins were significantly different in different lung tissues.The positive expression rates of Twist protein in non-small cell lung adenocarcinoma tissues and paracancerous tissues were 70.7%(29/41)and 21.9%,respectively(9/41),its expression rate in cancer tissues was significantly higher than that in paracancerous tissues(P=0.000),and the positive expression rate of N-cadherin protein in non-small cell lung adenocarcinoma tissues and adjacent tissues was 73.1%(30/41)and19.5%(8/41),respectively,were significantly higher in tumor tissues than in adjacent tissues(P=0.000).The transcriptional levels of Twist and N-cadherinwere basically consistent with the translational levels.In studies related to clinical pathology,it was found that the level of Twist and N-cadherin protein expression was significantly associated with lymph node metastasis and TNM stage(P < 0.05),and the patient’s gender,age and pathological differentiation were No correlation(P>0.05).Twist and N-cadherin expression were also positively correlated(r=+0.560,P=0.000);follow-up results showed survival from 4 months to 36 months,median survival time was 24.5 months,Kaplan-Meier survival analysis The positive survival rate of Twist was lower than that of negative group(P<0.05).Cox regression analysis showed that TNM staging and lymph node metastasis could be independent prognostic factors of non-small cell lung adenocarcinoma.Conclusion: The expression of Twist and N-cadherin was significantly higher in cancer tissues than in adjacent tissues and benign lesions.The expression of Twist and N-cadherin was closely related to the clinical stage and lymph node metastasis of non-small cell lung adenocarcinoma.Twist may participate in the EMT process and affect the expression of N-cadherin.And then play a role in the process of metastasis and invasion of non-small cell lung adenocarcinoma.The high expression of Twist and the high expression of N-cadherin may be closely related to the occurrence,development,invasion and metastasis of non-small cell lung adenocarcinoma.It is of great value in the diagnosis and evaluation of non-small cell lung adenocarcinoma,and it is possible that It can be used as a potential biological target in the diagnosis and treatment of patients with non-small cell lung adenocarcinoma.The high expression of Twist and the high expression of N-cadherin are positively correlated with the expression of non-small cell lung adenocarcinoma.Twist may participate in the invasion and metastasis of non-small cell lung adenocarcinomaby participating in the EMT pathway to up-regulate the N-cadherin pathway.Wait for the process.
