Objective:This study aimed to detect the expression level of microRNA-3656 in the serum of lung cancer patients and normal healthy people by real-time fluorescence quantitative PCR method,analyze the expression characteristics of microRNA-3656,and explore its correlation with clinical parameters of lung cancer patients.,providing some value for the early diagnosis,treatment and prognosis of lung cancer.Methods:To collect early morning fasting blood samples from 70 patients with lung cancer who were admitted to the Oncology Center of the Second Affiliated Hospital of Xiangya College of Medicine,Central South University and Department of Oncology of Nanhua Hospital Affiliated to Nanhua University from July 2015 to March 2016 and 38 healthy people from Nanhua Hospital Affiliated Nanhua University.Real-time fluorescent quantitative PCR(qRT-PCR)was used to detect the expression level of microRNA-3656 in the serum of lung cancer group and healthy person group.The t-test was used to analyze the measurement data.Chi-Square was used to analyze the relationship between microRNA expression level and clinical data of lung cancer patients.The efficacy of MicroRNA-3656 in the diagnosis of lung cancer was analyzed using receiver operating characteristic(ROC)curves to obtain the area under the ROC curve(AUC)values.The mean expression level of MicroRNA-3656 was calculated based on 2-?? Ct,and divided into high-expression group and low-expression group.The overall survival period of high-expression group and low-expression group in lung cancer group was analyzed by Kaplan-Meier method.Results:1.The expression level of microRNA-3656 in the serum of patients with lung cancer was significantly higher than that in the normal group(P<0.01).2.In the lung cancer patient group,Chi-Square test analysis showed that the expression of microRNA-3656 was significantly associated with clinical stage and distant metastasis(P<0.001),and was not related to gender,age,pathological type(P>0.05).3.The plasma microRNA-3656 expression level affected the survival time of patients.The median survival time of patients with high microRNA-3656 expression(n=21,11.38 ±0.98)was significantly shorter in patients with low expression(n=41,24 ±0.86).(P<0.001);4.Analysis of the diagnostic efficacy of microRNA-3656 by ROC curve,microRNA-3656 has a certain diagnostic value for lung cancer(P<0.05).Conclusions:1.MicroRNA-3656 shows abnormal high expression in serum of patients with lung cancer,and detection of microRNA-3656 in human serum is expected to be used for the early diagnosis of lung cancer.2.The expression of microRNA-3656 is closely related to tumor stage and metastasis,and it is related to the survival time of lung cancer patients.This suggests that microRNA-3656 is expected to be used for the prognosis of lung cancer patients.
