Background: Depression is a neuropsychiatric disease caused by genetic factors and environmental factors.Clinical manifestations include decreased energy,depressed mood,cognitive impairment,and even severe suicide.Although the research on depression is more and more in-depth,there is no ideal antidepressant drug,and long-term use of synthetic antidepressants will cause many adverse reactions.Recently,the treatment of depression by Chinese medicine has received more and more attention from clinicians and researchers.Tanshinone ?A has anti-inflammatory,anti-oxidation,and anti-apoptosis functions and is commonly used in the treatment of cardiovascular and cerebrovascular diseases.Studies have shown that tanshinone ?A has a significant protective effect on Alzheimer's disease and cerebral ischemia;however,it is unclear whether tanshinone ?A has an antidepressant effect.This topic mainly studies the antidepressant effect of tanshinone ?A on primary depression and its mechanism.Merhods: Experimental animals: Limiting spatial behavior,using TST and FST to detect mice exhibiting depression-like behavior;Protein detection: Western,Q-PCR.Results: ICR mice were subjected to three weeks of spatial behavioral restraint,and TST and FST detected depression-like behavior in mice;after tanshinone ?A treatment,Tanshinone ?A can significantly reduce the immobility time of mice in TST and FST,suggesting that tanshinone ?A has antidepressant effect.The expression of ERK-CREB-BDNF signal pathway related protein in the brain tissue of depressive model of tanshinone ?A and the stress model induced by dexamethasone was significantly increased,and the expression level of BDNF mRNA in mice after tanshinone ?A treatment was also significantly increased.However,the administration of ERK inhibitor SL327 significantly reduced the expression of ERK-CREB-BDNF signaling pathway-associated proteins and blocked the antidepressant effect of tanshinone ?A.Conclusions: Studies have shown that tanshinone ?A has good antidepressant effect,and its antidepressant mechanism is activated by activating ERK-CREB-BDNF signaling pathway,providing a theoretical basis for the clinical treatment of depression in tanshinone ?A,due to tanshinone ?A With less side effects,it is hopeful to become a highly effective drug for clinical treatment of depression.
