Objective : To investigate the effect of fibrin-derived peptide B?15-42 and lumbrukinase EFEa on atherosclerosisMethods:New Zealand rabbits were given the high-cholesterol diet for 8 weeks to establish the animal model of atherosclerosis.They were divided into four groups: the control group,the fibrin-derived peptide B?15-42 group,the lumbrukinase EFEa group and the rosuvastatin group.First test the hematological differences in each group.Subsequently,the aortic tissue was taken for detecting the content of fibrinogen in the vascular tissue by immunoblotting.Paraffin sections were made from aortic tissue.Fibrinogen antibody and CD68 antibody were used to observe the intima.The area of atherosclerosis was observed by HE staining.Finally,cut open the aortic tilapia to observe the area of atherosclerosis on the vascular by oil red staining.Results:Hematology,Serum lipid levels decreased in the statin group.There was no significant difference in the levels of fibrinogen in each group.The ICAM-1 in each intervention group and MIF of B?15-42 groups decreased compared with the control group.Fibrinogen immunohistochemical staining and Western blotting experiments found that fibrinogen in atherosclerotic tissue was significantly deposited in the control group with the highest content and decreased after the drug intervention.CD68 antibody immunohistochemical markers of inflammatory cells found that intervention group intimal infiltration of inflammatory cells decreased.For animal tissue examination,B?15-42 group atherosclerosis tissue area compared with the high-fat diet decreased,and lumbrokinase EFEa and rosuvastatin group atherosclerosis has more reduction.Conclusion:The fibrin-derived peptide B?15-42 can effectively inhibit the aortic atherosclerosis of high cholesterol-diet New Zealand rabbits by protecting endothelial function,reducing local fibrinogen deposition,decreasing the level of atherosclerosis-related inflammatory factors,and infiltrating inflammatory cells in the aorta.The lumbrokinase EFEa has fibrin-derived peptide B?15-42-like antiatherosclerotic effects.
