Purpose: The study is to explore the the expression and correlation of GOLPH3 and mTOR protein in patients with local advanced oesophageal squamous cell carcinoma,and to analyze the relationship between the prognosis of these patients with radical radiotherapy or chemoradiotherapy.Methods: We retrospectively reviewed the records of patients with local advanced oesophageal squamous cell carcinoma at the Shandong Cancer Hospital and Institute between January 2011 and December 2015.Immunohistochemistry was used to assess GOLPH3 and mTOR(mammalian target of rapamycin)in tumor tissues.The Kaplan–Meier method compare the overall survival(OS)and progression-free survival(PFS),and the Log-rank test was used to compare the difference between groups.Results: In this study,a total of 179 patients with local advanced oesophageal cancer with radical radiotherapy or chemoradiotherapy were collected,and 65 cases were analyzed by immunohistochemistry.Among them,the expression of GOLPH3 was different in patients' progression(P=0.033).Immunohistochemical results showed that the positive expression rates of GOLPH3 and mTOR were 35.4%(23/65)and 47.7%(31/65),respectively.The expression of GOLPH3 was correlated with mTOR.The difference was statistically significance(P<0.05),and Pearson's contact number rp = 0.413.Survival analysis showed that the median survival time of GOLPH3 patients was only 23.3 months,while the low-expression group was 33.1months.The median survival time of mTOR high expression patients was only 27.2months,while the low expression group was 37.2 months(P<0.05).Conclusion: High Expression of GOLPH3 in local advanced Oesophageal Squamous Cell Carcinoma Correlates with Poor Prognosis.And the expression of GOLPH3 was high correlated with mTOR.Considering that GOLPH3 may activate the mTOR signaling pathway and increase the sensitivity of rapamycin in tumor cells,it provides important insights into individualized treatment of patients in the future.
