Objective:In recent years,more and more researchers pay attention to the attenuation and synergistic effect of traditional Chinese medicine?TCM?,fuzi is one of the most common traditional Chinese medicines,due to its remarkable curative effect and side effects,it has attracted much attentionin natural medicine community.Attenuation of the side effects and improvement of synergistic effects of Fuzi are the first priority for its clinical applications.Poria cocos,also as a common TCM and an edible and pharmaceutical mushroom,it is widely used in clinic due to its pharmacological effects includinglighting seepage and removing dampness,tonifying spleen and nourishing heart,and so on.In this study,the attenuating and synergistic effect and their compatibility law of Poria cocos combined with Hei-shun-pian?HSP?,a TCM listed in Chinese Pharmacopoeia,was investigated,to provide a basis for further studies on complex prescription compatibility law of TCMs.Methods:Based on the experimental guidelines of acute toxicity of TCM and natural medicine,the acute toxicity as the macroscopic comparison of different components of HSP on normal mice was carried out,based on the results,acute toxicity as the macroscopic comparison and micro-related biochemical indicators LDH,AST,ALT,BUN,CRE also were tested,study the attenuation of Poria cocos combined with HSP.Secondly,chronic heart failure model was established by ligation of left coronary artery and reducing diet method.The pathological changes of heart were observed by HE staining and the biochemical indexes LDH,CK,Ang-?,AT1R related to cardiac function were detected.Subcutaneous injection of 1%carrageenan the paw swelling model was used to analyze the anti-inflammatory effect of paw swelling index.The physical pain model was established by hot plate method.The chemical pain model was established by writhing acetic acid method,the reaction time,the number of writhing reaction and writhing inhibition rate as an indicator to initially understand the analgesic effect of different proportions of Poria cocos combined with HSP,and inspect the pharmacodynamic effect so that study the synergistic effects of Poria cocos combined with HSP.Results:The LD500 and 95%confidence limit of alcohol extracted components were278.0 g·kg-1·d-11 and 265.5291.0 g·kg-1·d-11 respectively,The MLD of all-components was 24.0 g·kg-1·d-11 and the MLD of water extracted components was 160.0 g·kg-1·d-1,before and after the compatibility of HSP,in the normal mice acute toxicity test,HSP alcohol group died two mice,Restlessness immediately,followed by nausea salivation,hyperspasmia,diarrhea,convulsions and prostrate motionlessness were the main acute toxical symptoms and signs caused by HSP,most of which occurred within 8.0 h after gavage administration.Different proportions of Poria cocos combined with HSP alcohol group their own weight growth rates vary greatly.In the LDH showed that HSP alcohol group compared with the control group?P<0.05?,with significant differences,and other indicators compared with the control group?P>0.05?,with no significant difference,however,LDH,AST,ALT,BUN and CRE in different ratios of Poria cocos combined with HSP decreased with the increase of compatibility ratio.The better attenuation effect of HSP combined with Poria cocos was 1:1 and 1:2.In the cardiac,anti-inflammatory analgesic experiments,the effect of different proportion of Poria cocos combined with HSP alcohol groups were better than that of water groups,and Poria cocos could improve the therapeutic effect of HSP on CHF SD rats,when compatibility of HSP and Poria cocos ratio in 1:1 and 1:2,the synergistic effect was better;Poria cocos also could enhance the anti-inflammatory effect,with the Poria cocos compatibility increased,the inhibitory effect of rat paw swelling have increased trend,and the HSP:Poria cocos 1:2 alcohol extract group was the most obvious inhibition;HSP:Poria cocos 1:1 alcohol extract group in the Chinese medicine extract groups was the least number of writhing,and the inhibition rate to 38.52%,close to the positive control group,and HSP:Poria cocos 1:1 alcohol group basal threshold of pain in the traditional Chinese medicine extract groups also increased the most.Conclusion:This study provided simple and systematic experimental evidence for the attenuation and synergistic effects of Poria cocos combined with HSP and the better compatibility ratios are 1:1and 2:1,the compatibility ratios are similar to the classic prescriptions which included Poria cocos and HSP simultaneously.Therefore,there is the regularity could be found when the ancients took advantage of traditional Chinese medicine,prescriptions compatibility principle also were scientifically based.These findings could help better understand the compatibility regularity of Poria cocos and HSP in clinical,and also provide the experimental basis for the rational use of Poria cocos and HSP.
