Objective:Through the establishment of the chronic myocardial ischemic model by injecting isoproterenol(ISO)drugs, investigate the protective effects of Poria Cocos(PC) and Cinnamomum cassia(CC) on chronic myocardial ischemia. And identify biomarkers in serum,at the same time investigate metabolic pathways;Methods: The chronic myocardial ischemia model requires small doses and prolonged control. 48 SD rats were divided into 6 groups randomly, set PC-CC group, PC group,CC group, model group, normal group, control group. Then after 28 days, changes of electrocardiogram ST segment, degree of lactate dehydrogenase(LDH), creatine kinase(CK)and(CK-MB) and HE staining of the heart tissue of rats were observed. Atlast,the characteristics of metabolites in rats in serum were measured and investigate the metabolic pathways of serum by nuclear magnetic resonance method,principal component analysis and orthogonal partial least squares discriminate analysis.Result: Compared with normal group, ST segment voltage each group rises(P <0.01), suggesting the model is accurate. After the treatment, compared with model group, ST segment voltage is lower, there are significant differences; Compared with normal group, the model group level of LDH, CK, CK-MB increased significantly in serum,with significant difference(P < 0.01), suggesting model accurate; After the treatment, PC group,CCgroup, PC-CC group effectively reduce the content of serum LDH, CK, CK-MB, and PC-CC drugs reduce the degree of the three levels lower,(P <0.01), there are significant differences; Compared with normal group, the edema degree of model group increases and the arrangement is not neat, texture unclear. Through the treatment, PC, CC, PC-CC cell edema degree is reduced gradually. And the cell neat,texture clear; Under electron microscope,compared with normal group, model group mitochondria are swelling, myocardial tissue cells scattered.After the gavage,poria cocos, cassia twig, poria cocos, cassia twig group cells gradually neat, mitochondria structure complete, intercalated complete.29 metabolites are detected, including Carbohydrate, amino acids, organic acids and so on. PC-CC drugs induced some physical changes in rats, but also changed to the metabolic disorders and amino acid metabolism disorders, maintain metabolic balance through the changes of energy metabolism and regulating metabolism of amino acids,Through the unsupervised principal component analysis, all groups of samples are limited within the confidence interval, with no abnormal samples.Model group and normal group, PC group, CC group and PC-CC group samples separate trend is obvious.Compared with pc group, CC group, PC-CC group sample are closer to the normal group;Conclusion: Compared with single drugs, PC-CC drugs can change the degree of myocardial ischemia more effectively.Many metabolic pathways, such as the course of Krebs cycle, glycolysis, choline metabolism, amino acid metabolism involved in serum.
