The Qi Teng Xiao Zhuo Granule Treated The Chronic Glomeriulonephritis(Cgn) Rats Based On LBP/CD14/FOS Signaling Pathway:A Mechanism Exploration

Objective To study the effect of Qiteng Xiao Zhuo Granule on LBP/CD14/FOS signaling pathway in model of chronic glomerulonephritis?CGN?rats.MethodsSD rats were randomly divided into six groups,including normal group,model group,Qiteng Xiao Zhuo Granule?21.6g/kg?10.8g/kg?5.4g/kg?,and Huangkui capsule(1.0g·kg^-1).Except for nomal control group,the rest groups were given adriamycin induced CGN rats,which was intravenously injected into the tail of rat in the rest groups.The rest groups,including Qi Teng Xiao Zhuo granule and Huangkui capsule groups,intravenously injected adriamycin two times?n=8?;adriamycin(3.5 mg·kg^-1)was to be injected on the first day and adriamycin(3.0mg·kg^-1)was to be injected on the fourteenth day.On the 21st day,Rats urine was collected for 24 hours in the metabolism cages,in order to check 24-hour urinary protein quantitation.24-hour urinary protein quantitation more than 50 mg/kg,which indicated model replication successful.After model establishment,the Qi Teng Xiao Zhuo granule group were given a gastric gavage of Qiteng Xiao Zhuo Granule?21.6g/kg?10.8g/kg?5.4g/kg?and Huangkui capsule group were given Huangkui capsule(1.0g·kg^-1),once per day for 30 days.The control normal group and model group were given distilled water.After the final dose,all the rats were put into metabolism cages and urine was collected for 24 hours.Pathological changes were observed by Hematein Eosin?HE?;The RT-PCR?real-time fluorescent quantitative PCR?was performed to detect the expressions of LBP?CD14?JNK?P38 mRNA,and Western blotting to detect the expressions of LBP?CD14?p-JNK?p-P38 protein.ResultsCompared with model group,the Qiteng Xiao Zhuo Granule not only effectively decreased expressions of LBP?CD14?JNK?P38 mRNA,but also inhibited expressions of LBP?CD14?p-JNK?p-P38 in the kidney.ConclusionThe Qiteng Xiao Zhuo Granule has certain treatment function on model of CGN rats,which might be related to decrease the expressions ofLBP?CD14?JNK and P38 in the ckidney and block LBP/CD14/FOS signaling pathway.
