PurposesTongue squamous cell carcinoma(TSCC)is well-known for its high rate of proliferation,recurrence and nodal metastasis.It is reported that BTBD7 may serve as a predictor of metastasis and a potential therapeutic target for non-small cell lung cancer(NSCLC)through regulating E-cadherin.However,whether BTBD7 participates in the metastasis and affects the prognosis of TSCC has not been identified.In this article,we researched the BTBD7 expression in TSCC and correlation with E-cadherin.In vitro experiments,siRNA targeting BTBD7 was employed to investigate the effect of BTBD7 on the biological behavior of TSCC and its possible mechanisms,to provide a basis for predicting the prognosis.MethodsWe used Immunohistochemical staining to study the expression of BTBD7 and E-cadherin in 53 TSCC tissue and 20 adjacent normal tissue to identify biomarkers for malignancy.BTBD7 was positive at cytoplasm,while E-cadherin was positive at cell membrane and cytoplasm.BTBD7 was positive for brown and granularsubstance in cytoplasm,and E-cadherin was positive for brown granules in cell membrane.SPSS 23 software was used for statistical analysis.Chi square test and Fishers exact test were used to examine the difference in expression of BTBD7 and E-cadherin in TSCC and adjacent normal tissues and their relationship with pathological parameters.Spearman correlation analysis was used to examine the relationship between BTBD7 and E-cadherin expression in TSCC.In vitro experiments,siRNA targeting BTBD7 was employed to investigate the proliferation,invasion and migration ability of Cal-27,and detect the expression of E-cadherin from RNA and protein levels,to further explore the mechanism of BTBD7 in the biological behavior of tongue cancer cells.ResultsTotal positive rate of BTBD7 expression was 41.51%in TSCC to 10%in normal tissues comparatively.The expression was significantly associated with the lymphatic metastasis,differentiated degree and distant metastasis(P<0.05)but no correlation with gender,age,and tumor size(P>0.05).Total positive rate of E-cadherin expression was 64.15%in TSCC to 100%in normal tissues comparatively.The expression was significantly associated with the lymphatic metastasis,differentiated degree and distant metastasis(P<0.05)but no correlation with gender,age,and tumor size(P>0.05).The results of Spearman correlation analysis showed that the expression of BTBD7 and E-cadherin in the tongue cancer tissues was negatively correlated(rs=-0.392,P=0.04),indicating that the expression of E-cadherin decreased gradually with the increase of BTBD7 expression.Downregulation of BTBD7 expression in tongue cancer cells by siRNA significantly lead to inhibit cancer cell invasion and effectively restore E-cadherin expression.ConclusionsCompared with normal tissue,BTBD7 showed high expression in tongue squamous cell carcinoma,while E-cadherin showed low expression.Both.of them have correlation with lymphatic metastasis,differentiated degree and distant metastasis.it showed a negative relationship between the expression of BTBD7 and E-cadherin in tongue squamous cell carcinoma(rs=-0.392,P=0.04).The downregulation of BTBD7 may be used as the upstream activation to induce EMT,promoting the expression of E-cadherin,participating in the development of tumor by regulating the movement of cancer cells,and affecting the clinical prognosis of the patients.BTBD7 may serve as a predictor of metastasis and a potential therapeutic target for TSCC.
