Malignant tumor can invade neighboring tissues or metastasis to the far-end of organs to invade and destroy the body,leading to death.The main features of Carcinogenesis related to normal cell cycle regulation and apoptosis which are out of control.Because all kinds of cancers development fast,most cancer patients were diagnosed with terminal cancer of which the treatments were difficult,and the results of the prognosis were not ideal,based on the clinical experience.Therefore,the cure of cancer was a big problem in medical profession.In the modern tumor medical treatment,chemical medication has very important role.Most chemotherapy drugs in clinical medicine served as first-line use at present.However,these chemotherapy drugs had inhibitory or cellulotoxic effect to both tumor cells and normal cells,leading to the serious side effects in patients who usually had to use them to continue life,which had not fundamentally improved the life quality.Along with the rapid development of the molecular biology and the genetic engineering as well as the intensive study related to molecular mechanisms of tumorigenesis,the development of the antitumor drugs has entered into a new stage,in which biotechnology antitumor drugs were appeared.They were targeted and had more targets,less adverse reactions and better curative effect,which made them one of the fastest development directions of antitumor drugs,for example monoclonal antibody.Peptide mapping method is very important for monoclonal antibody identify testing.The method was established for peptide mapping of recombinant monoclonal antibody pharmaceuticals in denaturing conditions,reducing reagent was added to recombinant antibody solution,all the crosslinked disulfide bonds were broken,and then alkylation reagent was added to block the free sulfhydryl residues,the resultant antibody molecules stayed in the solution as dissociated and extensively stretched peptide chains.The trypsin was added to the solution and the antibody was digested to small peptide fragments.Reversed phase HPLC was used to separate e the peptide fragments.The method for peptide mapping method was effective in proteins such as recombinant antibodies with complex structure.It can used in identify test for monoclonal antibody.
