Objective To prepara anti-SMO(human Spermine oxidase)monoclonal antibody andto analyze its titer, antigen specifieity, species specifieity and usability in Western blotting,Immunoprecipitation, Immunohistochemical analysis, Immunofluorescence, Flowcytometry (FCM) and other technique.Methods SMO recombinant protein with6×His tag was expressed in BL21(DE3)and purified by Ni-NTA resin. The purified recombinant SMO was used to immunizeBalb/C mouse and the anti-SMO monoclonal antibody was acquired by hybridomatechnique. The titer and antigen specificity of this antibody were identified by ELISA,western blot and immunohistochemical analysis.Results A hybridoma cell line which could stably secret anti-SMO mAb was obtainedsuccessfully. This mAb can be used in ELISA, Western blotting, Immunoprecipitation,Immunohistochemical analysis, Immunofluorescence, Flow cytometry (FCM) for SMOassay.Conclusion The mouse against human SMO monoelonal antibody with high titer andantigen specificity was obtained successfully, which lays a foundation for SMO-relatedmolecule biological, pathological and anti-tumor researches.
