Experimental Study And Clinical Observation Of Sildenafil Of Liuweibuqi Capsules In JAK/STAT Pathway Of Alveolar Epithelial Cells Intervened By CSE+LPS

?Objective?Traditional Chinese medicine has unique advantages in the treatment of COPD.According to the theory of replenishing lung qi in traditional Chinese medicine,we developed the Chinese medicine Liuweibuqi Capsule.Based on the previous study of Liuweibuqi capsules in the treatment of chronic obstructive pulmonary disease,the mechanism of the treatment of COPD deficiency of lung qi was further discussed.In this study,“Pulmonary Qi and Division Respiratory” is used as a theoretical guide for traditional Chinese medicine.SiRNA interference technology was used to silence STAT gene in alveolar epithelial cells after cigarette smoke and lipopolysaccharide intervened,and the Chinese herbal compound Liuweibuqi capsule was used to analyze the JAK/STAT pathway.The regulatory mechanisms,and simultaneously observed changes in the clinical patient-related indicators,to explore the role of Chinese medicine clinical prevention and treatment of COPD.?Methods?1.24 SD rats were selected and were given different doses of Liuweibuqi capsules by intragastric administration,and normal control groups were set up to make low,middle and high proportions of drug-containing serum.The primary alveolar epithelial cells were cultured and the models were established.After CSE+LPS intervention was successful,drug-containing serum was added to detect the expression of related inflammatory factors and proteins.2.Twenty patients with lung qi deficiency from the Department of Respiratory Department of Anhui University of Traditional Chinese Medicine and wards of COPD during the stable period were randomly divided into observation group and control group with 10 cases in each group.Placebo and Liuweibuqi capsules were given orally respectively,and the total observation time period was 3 months.Blood samples were taken before and 3 months after treatment,and the lung function indices FEV1,FVC and FEV1/FVC,6-minute walking distance,and serum JAK1,JAK2,STAT1,STAT3,and STAT5 before and after treatment were observed.3.Using SPSS16.0 software,according to the observed indicators and the data obtained for statistical processing.?Results?1.The results of cell experiments showed that Liuweibuqi capsule-containing serum can increase the viability of model cells and reduce apoptosis.In addition,the levels of IFN?,IL-1?,IL-6,TNF-? and TGF-?1 in cells after 24 hours of CSE+LPS treatment can be reduced,and the expression of these factors is dose-dependently changed with the proportion of serum-containing drugs.The expression level of IL-10 is contrary to the above indicators.In addition,JAK1,JAK2,STAT1,STAT3 and STAT5 and their phosphorylation were also significantly reduced in the cells.Compared with each group,P<0.01 or P<0.05,the difference was statistically significant.2.After knocking down the STAT3 gene by SiRNA,the detection of various indicators in the model cells revealed that the expression of IL-10 was enhanced and the level of other inflammatory markers was decreased.In addition,the silencing of STAT3 reduced the CSE+LPS-induced inflammatory cytokines.Release,promotes cell viability.3.In the clinical trials,after treatment,the lung function index in the Liuweibuqi capsule group were improved compared with the pretreatment and control groups.After treatment,the serum JAK1,JAK2,STAT1,STAT3,and STAT5 were improved.The index also decreased significantly,compared with the control group,P<0.05,with statistical significance.?Conclusion?The underlying pathogenesis of COPD deficiency of lung syndrome is "lung loss,decline,and segmental disturbance".Overactivation of the JAK/STAT pathway is an important factor in the progressive decrease of lung function of COPD.Silencing of STAT gene by siRNA can improve the inflammatory response of COPD.Chinese herbal compound Liuwei Buqi capsules can interfere with the JAK/STAT pathway,block or inhibit the activation of the JAK/STAT pathway,regulate the body's inflammatory response,regulate siRNA expression and cytokine network,so that the lungs of the reconstructive function,and ultimately improve the COPD Lung Qi deficiency respiratory system function.
