Objective: First,from the m RNA level and protein level to study whether the expression of B7 family members is related to gastric cancer,to explore whether it can serve as a potential target for gastric cancer immunotherapy.Second.To investigate the correlation between B7 family member protein expression and clinical indicators of gastric cancerMethods: First,the operating room collection Rocky Mountain Hospital March 2016-December 2016 76 cases of freshly resected specimens of gastric cancer surgery,take the cancerous tissue as the experimental group and the normal tissue next to the cancer tissue edge 5cm above the normal stomach tissue as a control group.The specimen was divided into two parts after isolation.A part(about 600 mg)was placed in a 5 ml EP tube containing RNA protection solution immediately after being isolated and stored at room temperature for detection of gastric cancer(experimental group)and adjacent tissues by Q-PCR(control).The expression level of 10 members of the B7 family in the group)was compared with that of the t-test.Second.The pathology department collected paraffin immunohistochemistry for paraffin blocks of 160 patients with gastric cancer from 2011 to 2012 in our hospital.The chi-square test analyzed B7-1,B7-2,B7-DC,B7-H2,B7-H3,B7.-The correlation of H6,B7-H7 protein expression with clinical parameters(age,sex,depth of invasion,lymph node metastasis,differentiation,tumor size,stage,survival time).Result:1 Immunohistochemical results 1.1B7-1 protein expression is localized to the cell membrane and cytoplasm in the gastric gland.The expression of B7-1 in gastric cancer tissues was significantly lower than that in adjacent normal tissues,and the difference was statistically significant(P<0.05).No correlation with patient survival time.1.2B7-2 protein expression is localized to the cell membrane and cytoplasm in the gastric gland.The expression of B7-2 in gastric cancer tissues and normal tissues around the tumor were highly expressed,and the difference was not statistically significant(p>0.05).There was no correlation between the expression level of B7-2 and the patient’s age,depth of tumor invasion,degree of tumor differentiation,lymph node metastasis,tumor stage,tumor size,clinicopathological parameters,and there was a certain correlation with gender.Male patients were in gastric cancer tissues.B7-2 protein expression was higher than that of female patients(P<0.05).There is also no correlation with patient survival time.1.3 B7-H2 protein expression is localized to the cell membrane and cytoplasm in the gastric gland.The expression of B7-H2 in gastric cancer tissues and adjacent normal tissues was high,and the difference was not statistically significant(P>0.05).There was no correlation between the expression level of B7-H2 and the patient’s age,sex,depth of tumor invasion,tumor differentiation,lymph node metastasis,tumor clinical stage,and tumor size clinical pathological parameters,ie different age,gender,depth of tumor invasion,There was no significant difference in the expression level of B7-H2 between samples with different degrees of tumor differentiation,lymph node metastasis,clinical stage,and tumor size,and there was no correlation with the survival time of patients.1.4 B7-H3 protein expression is localized in the cell membrane and cytoplasm of gastric glands.The expression of B7-H3 in gastric cancer tissues was significantly higher than that in adjacent normal tissues,and the difference was statistically significant(P<0.05).There was no correlation between the expression level of B7-H3 and the patient’s age,sex,depth of tumor invasion,tumor differentiation,lymph node metastasis,clinical stage parameters of tumor clinical staging,ie different age,gender,depth of tumor invasion,degree of tumor differentiation,There was no significant difference in the expression level of B7-H3 between samples with lymph node metastasis and different clinical stages.Correlation with the size of the foci,the greater the foci(> 5cm),the more B7-H3 protein expression,and no correlation with survival time.1.5 The expression of 1.5B7-H6 protein was localized in the cell membrane and cytoplasm of gastric glands.The expression of B7-H6 in gastric cancer tissues was significantly lower than that in adjacent normal tissues,and the difference was statistically significant(P<0.05).There was no correlation between the expression level of B7-H6 and the patient’s age,sex,depth of tumor invasion,tumor differentiation,lymph node metastasis,tumor clinical stage,tumor size clinical pathological parameters,ie different age,gender,tumor invasion depth,There was no significant difference in the degrees of tumor differentiation,lymph node metastasis,clinical stage,and tumor size.The survival time of B7-DC negative patients was longer(P<0.05).2 Q-PCR experiment results Relative expression levels of genes were calculated using comparative CT values.The formula was: F=2-ΔΔCT,ΔΔCT value =(CT value of gastric cancer group target gene-CT value of internal reference gene in gastric cancer group)-(CT value of adjacent cancer group-cancer adjacent Group internal reference gene CT value).The experimental results showed that the m RNA expression of B7-1: cancer tissue was significantly less than that of adjacent normal tissues(p<0.05).B7-2: There was no significant difference in m RNA expression between cancer tissues and adjacent tissues.B7-DC: The m RNA expression level in the adjacent tissue was significantly higher than that in the cancer tissue(p<0.05).B7-H1: There was no significant difference in m RNA expression between cancer tissues and adjacent tissues.B7-H2: There was no significant difference in m RNA expression between cancer tissues and adjacent tissues.B7-H3: The m RNA expression level of cancer tissue was significantly higher than that of adjacent normal tissues(p<0.01).B7-H4: There was no significant difference in m RNA expression between cancer tissues and adjacent tissues.B7-H5: There was no significant difference in m RNA expression between cancer tissues and adjacent tissues.B7-H6: The m RNA expression in the adjacent tissue was significantly higher than that in the cancer tissue(p<0.05).B7-H7: There was no significant difference in m RNA expression between cancer tissues and adjacent tissues.3 Concordance analysis of m RNA and protein expression in B7 family members The expression of B7 family member proteins and m RNA in 7 immunohistochemical experiments was almost identical in cancer tissues and adjacent tissues.Conclusion: B7-1,B7-DC,B7-H6,and B7-H3 may be related to gastric cancer and may be potential targets for the treatment of gastric cancer.The expression of B7-2 in sex was different.The expression of male cancer tissue was higher than that of female.The larger the B7-H3 was,the larger the foci were in cancer tissues.The expression of B7-DC was maybe correlated with the survival time of patients.
