Object: To study the effect of Black Chinese wolfberry on the expression of TNF-? and Capase-3 in the ischemia-reperfusion of long-term high intensity training rats.Method: 60 SPF Wistar rats were divided randomly into three groups which are sedentary group(group A,12),long-term high intensity training group(group B,24)and Black Chinese wolfberry+ long-term high intensity training group(group C,24)by their numbers.Group A did not carry out any exercise,Group B and group C were arranged to do mid-intensity swimming exercise during the first three weeks,and started to build up the model of overtraining from the beginning of the forth week.We collected the urine of all the rats every three weeks to detect the concentration of NGAL and confirmed weather the rats have carried on the sport-related renal injure.Collected the venous blood at the end of sixth week after the rats were killed 24 hours to observe serum creatinine,blood urea nitrogen concentration changes.At the same time,bilateral kidneys in rat were taken,one was used to detect the change of TNF-??Caspase-3 and the other was used to make tissue biopsies to detect the morphological structure.Result:(1)The concentration the serum creatinine,blood urea nitrogen and NGAL of B group and C group were significantly higher than A group.(2)The expression of TNF-? and Caspase-3 of B group were significantly higher than A group.(3)The C group's concentration the serum creatinine,blood urea nitrogen,NGAL,the expression of TNF-? and Caspase-3 is significantly lower than the B group.Conclusion:(1)we succeeded to build up the model of ischemia-reperfusion during the three-weeks training.(2)ischemia-reperfusion can enhance the expression of TNF-? and Caspase-3.(3)Black Chinese wolfberry have the ability to restrain the sport-related ischemia-reperfusion and the expression pf TNF-? and Caspase-3 in renal tissues.
