Objective:To study the correlation between CTRP1 expression and coronary atherosclerosis in different tissuesMethods:1.Detection of CTRP1 protein expression in plasma,subcutaneous adipose tissue and epicardial adipose tissue in patients with coronary heart disease:1.By coronary angiography,the patients were divided into coronary heart disease(CHD n=12)group,non-crown Non-coronary heart disease(NCHD n=30)and control groups(n=12).The CHD group refers to patients with coronary artery bypass grafts that have three branches of coronary artery disease,left main disease,or double-vessel disease including anterior descending coronary artery disease.The NCHD group refers to patients undergoing heart valve replacement or atrial(ventricular)septal repair.Patients older than 60 years old were excluded from patients with coronary artery disease based on coronary angiographic results.The control group was a normal person who performed a health examination at the physical examination center.Plasma CTRP1 was measured by enzyme immunosorbent assay(ELLSA).The expression of CTRP1 in adipose tissue was compared between immunohistochemistry and western blot(WB).All patients were calculated body mass index(BMI),liver function determination,renal function,blood lipids,blood glucose,BNP,hs-TnI,echocardiography2.Determination of coronary artery thrombosis CTRP1 protein expression in patients with coronary heart disease:Patients with coronary artery disease who underwent coronary thrombectomy were included in the experimental group(n=3).Patients with peripheral arterial embolism were control group(n=5).The expression of CTRP1 in thrombus tissue was compared between two groups by histochemistry and Western blotting.3.Detection of CTRP1 protein expression in myocardial tissue of myocardial infarction mice:Twenty-four 10-week-old male KM mice were divided into operation group(n=12)and sham operation group(n=12)for coronary artery ligation.The coronary arteries were ligated for a period of time.The surgical components were surgical 3 days(group A,n=6),21 days(group B,n=6),and the sham operation group was sham-operated 3 days(group C,n=6),The sham component was sham-operated 21 days(group D,n=6).Immunohistochemistry and Western blotting were used to compare the differences of CTRP1 protein expression in myocardial tissue of mice in each group.Result:1.Plasma CTRP1 level test results:Plasma CTRP1 levels in patients with CHD were significantly higher than those in NCHD group and control group(p<0.01).There was no significant difference in plasma CTRP1 expression levels between NCHD group and control group(p>0.05).2.Comparison of CTRP1 levels in different tissues of the same patient:CTRPl<cardiac adipose tissue CTRP1 in the subcutaneous adipose tissue of the CHD group(p<0.05);subcutaneous adipose tissue CTRP1 in the NCHD group<epicardial adipocyte CTRP1<myocardial tissue CTRP1(p<0.05).3.CTRP1 levels in different tissues of CHD group and NCHD group showed no significant difference in the expression of CTRP1 protein in subcutaneous adipose tissue of CHD compared with NCHD group(p>0.05).The expression level of CTRP1 protein in CHD epicardial adipose tissue was higher than that in NCHD group(p<0.05).4.Comparison of levels of CTRP1 in NCHD group,comparison of atrial septal defect group,ventricular septal defect group,mitral valve replacement group,and aortic valve replacement group.Serum,subcutaneous adipose tissue,and epicardium between different subgroups CTRP1 expression in adipose tissue and myocardial tissue was not statistically different(p>0.05).5.The expression of CTRP1 protein in myocardial tissue of mice with myocardial infarction 3 days was higher than that in sham-operated 3 days.The difference was statistically significant(p<0.05).The expression of CTRP1 protein in myocardial tissue of mice with myocardial infarction 21 days was higher than that of sham-operated 21 days.The difference was statistically significant(p<0.05).6.CTRP1 levels in coronary thrombosis and peripheral arterial thrombosis showed no CTRP1 expression in both thrombus tissues.7.The expression of CTRP1 in plasma of CHD group was positively correlated with plasma hs-CRP and patient SBP(p<0.05).hs-CRP in plasma was an independent factor for CTRP1(p<0.05).Conclusion:1.The plasma CTRP1 protein expression is elevated in patients with coronary heart disease.2.The expression of CTRP1 protein was increased in epicardial adipose tissue of patients with coronary heart disease.3.Myocardial infarction,CRTP1 protein expression levels in myocardial tissue increased.There was no CTRP1 expression in coronary thrombosis and peripheral arterial thrombosis.5.CTRP1 protein expression level is positively correlated with SBP.6.Plasma hs-CRP is an independent factor for CTRP1.
