Low-impedance is a feature that the acupuncture meridian was first studied and recognized,more and more researches suggest that the connective tissue is the material basis of the acupuncture meridian;Recent studies have also shown that the acupuncture meridian may be a tissue with high concentration of reactive oxygen species?ROS?.Collagen is the main component of connective tissue,so this thesis attempts to study the low-impedance mechanism of acupuncture meridian from the perspective of collagen,especially its characteristics effected by ROS.The main research included:the effect of compositional conditions on the impedance characteristics of collagen gel,the capacity of collagen gel to "transmit" superoxide anion,and the effect of collagen on fluorescence intensity of reactive oxygen species probe?DCFH-DA?to pilot study low-impedance mechanism of acupuncture meridian.The results are shown as followings:1.The effect of compositional conditions on the impedance characteristics of collagen gel found ?The influence of the concentration,pH and ionic strength of collagen solution was positively correlated with the impedance;?Different temperatures caused three-strand spiral structure of collagen to changes in different degrees,and the impedance value increases with its structure disorder,when the temperatures at 25? or 37?,the impedance value was the smallest;?Collagen showed low resistance treated with 0.8 ?M of hydrogen peroxide,and the rest concentration of the hydrogen peroxide had no obvious effect on the impedance of collagen.2.The experiment of collagen gel "transmitting" superoxide anion found that it could"transmit" superoxide free radicals significantly,but agar gel had no such effect in the method of hydroxy la mine hydrochloride,fluorescent detection and organic solvent.3.The effect of collagen on fluorescence intensity of reactive oxygen species probe?DCFH-DA?found ?Different types of collagen significantly improved the strength of the fluorescence with a dose-dependent manner,which is probably due to its special triple helix structure and functional group of the amino acid residues;?The fluorescence of the PBS and DCFH-DA system was transiently enhanced by the addition of?PPG?10?a mimetic peptide of collagen?;?The interaction between?PPG?10 and DCFH-DA showed that,on the one hand,DCFH-DA was under the action of?PPG?10,and promoted to be oxidized to produce fluorescent substances;on the other hand,DCFH-DA increased the hydrophobicity of?PPG?10,structure changes may be from single chain into random coil structure based on the analysis by HPLC-UV,HPLC-FL,HPLC-MS.In summary,these results suggest:?Collagen is beneficial to produce low-impedance phenomenon of acupuncture meridian in certain conditions;?Collagen may acts as a carrier in the "transmission" of superoxide anion free radical in acupuncture meridian system,the"transmission" of superoxide anion radicals in connective tissue may contributes to the low-impedance of the acupuncture meridian system;?Collagen may act as an electron transfer promoter in the acupuncture meridian system.Collagen has the characteristics of the above structure,so that it may have a close relationship with the low-impedance characteristics of acupuncture meridian.
