In recent years,the number of breast cancer patients increased year by year,its morbidity and mortality in women with malignant tumors in the second,and tumor invasion and metastasis is the leading cause of death in breast cancer patients.Looking for ways to regulate the invasion and metastasis of breast cancer is very important to reduce the mortality of breast cancer patients.OVOL2 is a fairly evolutionary transcription factor,and its relationship with embryonic development has been reported by many studies,but OVOL2 is not well known about the development of tumors.In the preliminary study,OVOL2 was able to inhibit the EMT progression of colorectal cancer by inhibiting the expression of SLUG by inhibiting the transcriptional activity of Wnt signaling pathway,thus inhibiting the invasion and metastasis of colorectal cancer.This has raised questions about whether OVOL2 can affect the invasion and metastasis of breast cancer.We found that OVOL2 overexpression significantly inhibited the invasion and metastasis of cells by Invasive Transwell experiments,scratches and analysis of the Oncomine database.The genetic evaluation of the Neve cell line group reminds us that OVOL2 may inhibit the invasion and metastasis of cells by inhibiting tumor cell EMT.We observed the change of EMT-related marker gene by immunoprecipitation,real-time quantitative PCR and Western blotting.The experimental results were confirmed by the combination of mouse model and confirmed that OVOL2 could inhibit the expression of EMT The EMT process of tumor cells inhibits tumor cell invasion and metastasis.In addition,we found that overexpression of OVOL2 in breast cancer cells could inhibit the activity of TGF-? signaling pathway and the expression of related genes(inhibition of Smad4 expression and up-regulation of TGF-?Smad7 expression)by fluorescence real-time PCR and Western blotting.Through the above results,we propose that OVOL2 can inhibit the invasion and metastasis of breast cancer by inhibiting the process of epithelial-mesenchymal transition.In this paper,we linked OVOL2 and EMT in breast cancer,and initially revealed the molecular mechanism of OVOL2 protein inhibiting the invasion and metastasis of breast cancer,and elucidated the mechanism of regulation of breast cancer through this mechanism.This finding is of great significance for the study of OVOL2 protein function and breast cancer.
