Background:Despite recent advances in therapy for advanced lung adenocarcino ma,there continues to be an unmet medical need for effective treatment of stag e Ⅲb/Ⅳ.The prognosis of lung cancer is still not optimistic. Objective:In orde r to supply some theory evidences for the targeted therapy in lung adenocarcin oma,we research the effective of Ovol2 gene to epithelial-mesenchymal transitio n (EMT) and the probable molecular mechanism of it; Methods:Firstly,Ovol2 Expression was evaluated by immunochemistry in Clinicopathological sections. Secondly, A549 cells were treated with control and Ovol2 overexpressioned b y lentivirus infection.To evaluate the effective of Ovol2 gene to EMT, Real-tim e PCR were performed to test the mRNA level of genes correlated to EMT; Western Blot were performed for the protein level of following makers:E-cadhe rin, N-cadherin, Vimentin ect. Lastly, rescue and Luciferase,chromatin immun oprecipitation assay were performed to explore the probable molecular mechanis m. Moreover, we tested the migration and invasion ability of A549 cells by Tr answell and Wound Healing experiment. Results:After treated with Ovol2 ove rexpressed, the expression level of E-cadherin is rised,while the expression leve 1 of N-cadherin、Vimentin and Twistl was declined not only in the mRNA lev el but also in the protein level. Whats more Ovol2 can directly bind in the sp ecific region of Twistl promoter and repress its expression. In additon,Wound Healing and Transwell indicate that the migration and invasion ability of A54 9 cells was weaked. Conclusion:Ovol2 gene can suppress the distant metastasi s ability and invasion ability of A549 cells by inhibiteing the EMT. Furtherly it is in the means of transcriptionally represses Twistl.
